摘  要:目的:探究小儿咽扁颗粒治疗急性咽炎的作用机制。方法:选取小儿咽扁颗粒中的8味中药作为研究对象,运用数据库配合文献查阅,以口服生物利用度和类药性为筛选的限定条件进行中药化学成分的筛选;运用Symmap数据库对化学成分进行靶点的寻找,在GeneCards、OMIM数据库搜索咽炎相关蛋白,与靶点进行映射,并通过映射找到小儿咽扁颗粒治疗急性咽炎的靶点;运用Cytoscape 3.5.1软件进行中药-化学成分网络、中药-化学成分-靶点网络、成分靶点蛋白质-蛋白质相互作用网络;运用Venny 2.1进一步研究方中君药金银花与射干治疗咽炎的作用机制;运用DAVID数据库对映射后靶标进行基因本体(GO)生物过程和京都基因和基因组百科全书(KEGG)富集分析。结果:筛选出97个化学成分,治疗咽炎的107个靶点,其中隐黄素(Kryptoxanthin)、MYST组蛋白乙酰转移酶(单核细胞白血病)4(MYST4)、三磷酸腺苷结合盒转运体A成员1(ABCA1)、视黄酸受体α(RARA)、与Runt相关转录因子1(RUNX1)为其核心靶点。方中金银花的靶点蛋白数占全方治疗咽炎总靶点蛋白数的41.5%。结论:小儿咽扁颗粒可能通过对药物的反应、对乙醇的反应、凋亡过程的负调控、转录过程的正调控、发挥治疗炎症的作用。其作用机制可能与肿瘤坏死因子(TNF),叉头框转录因子O(FoxO),磷脂酰肌醇3激酶/蛋白激酶B(PI3K/AKT),缺氧诱导因子1(HIF-1),T细胞受体等信号通路有关。其中Kryptoxanthin、MYST4、ABCA1、RARA、RUNX1可能是为其发挥药效作用的核心靶点。Objective:To explore the mechanism of Xiao'er Yanbian Granule in the treatment of acute pharyngitis.Methods:Eight medicinal herbs in Xiao'er Yanbian Granule were selected as the research objects,and the chemical components of the medicinal herbs were screened by databases and literature review,with oral bioavailability and drug-likeness as limited conditions.Symmap was used to search the targets of the chemical components,and the proteins related to pharyngitis were searched in GeneCards and Online Mendelian Inheritance in Man(OMIM)and then mapped with the targets.The targets of Xiao'er Yanbian Granule in the treatment of acute pharyngitis were obtained by mapping.Cytoscape 3.5.1 was used to construct the medicinal herb-chemical component network,medicinal herb-chemical component-target network and protein-protein interaction network.Venny 2.1 was employed to further study the mechanism of the sovereign medicinal herbs in Xiao'er Yanbian Granule,Flos Lonicerae Japonicae and Rhizoma Belamcandae,against pharyngitis.The Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis of the mapped targets were performed using DAVID.Results:A total of 97 chemical components were screened from Xiao'er Yanbian Granule,and 107 targets were screened for the treatment of pharyngitis,among which kryptoxanthin,MYST4,ABCA1,RARA and RUNX1 were the core targets.The number of target proteins of Flos Lonicerae Japonicae in Xiao'er Yanbian Granule accounted for 41.5% of the total target proteins of the prescription in the treatment of pharyngitis.Conclusion:Xiao'er Yanbian Granule may play an anti-inflammatory role through the reaction to drugs and alcohol,negative regulation of apoptosis and positive regulation of transcription.The mechanism may be related to TNF,FoxO,PI3K/Akt,HIF-1,T cell receptor and other signaling pathways,among which kryptoxanthin,MYST4,ABCA1,RARA and RUNX1 may be the core targets for the therapeutic effect of Xiao'er Yanbian Granule.
