摘 要:目的:观察补肾促孕方(BCR)对环磷酰胺(CTX)导致子宫内膜损伤的保护作用,并且分析其药效成分。方法:SD大鼠腹腔注射CTX后分组灌胃给药,计算各组大鼠子宫指数,检测血清雌二醇(E_(2))水平,子宫内膜厚度及子宫内膜腺体数目,观察胚胎着床情况。运用UHPLC-LTQ-Orbitrap MS技术检测BCR的化学成分,结合药效与文献分析其药效成分。结果:与正常对照组比较,CTX组大鼠子宫指数有下降的趋势,血清E_(2)水平显著下降(P<0.05),子宫内膜厚度显著变薄(P<0.05),子宫内膜腺体数目、总胚胎数目及活胚数目均有减少的趋势;与CTX组比较,BCR组大鼠子宫指数有升高的趋势,血清E_(2)水平显著升高(P<0.05),子宫内膜厚度显著增厚(P<0.05),子宫内膜腺体数目、总胚胎数目及活胚数目有增加的趋势。BCR中共鉴定出84种化学成分,其药效成分推测主要集中于黄酮类。结论:补肾促孕方能够改善子宫内膜容受性,其药效成分推测为槲皮素和山柰酚等。Objective: To observe the protective effect of Bushen Cuyun Recipe(BCR) on cyclophosphamide on endometrial damage and to explore the pharmacological material basis of BCR. Methods: SD rats were injected intraperitoneally with cyclophosphamide(CTX) and administered by gavage in groups. The uterine index was calculated, the serum estradiol(E_(2)) level was measured, the endometrial thickness and the number of endometrial glands were detected pathologically, and the fertilization experiment was carried out to observe the embryonic implantation, and finally, the chemical composition of BCR was detected by UHPLC-LTQ-Orbitrap MS technology, and its pharmacological material basis was analyzed by the pharmacological effects and the literature. Results: The animal experiments showed that compared with the control group, the uterine index of rats in the CTX group tended to decrease;the serum E_(2) level decreased significantly(P<0.05);the endometrial thickness thinned significantly(P<0.05), the number of endometrial glands, the total number of embryos and the number of live embryos of rats both tended to decrease;compared with the CTX group, the uterine index of rats in BCR group had a tendency to increase;the serum E_(2) level of rats increased significantly(P<0.05);the endometrial thickness of rats increased significantly(P<0.05), and the number of endometrial glands, the total number of embryos and the number of live embryos of rats both had a tendency to increase. The results of pharmacological material basis study showed that a total of 84 chemical components were identified from BCR, among them, flavonoids chemical drug substances were closely related. Conclusion:BCR can improve the endometrial receptivity, and its pharmacological material basis is presumed to be components such as quercetin and kaempferol.
