摘  要:目的:探索二至丸含药血清对自然衰老细胞模型水通道蛋白1(AQP1)及细胞膜通透性的影响。方法:二至丸含药血清干预成人真皮成纤维细胞自然衰老细胞模型,观察不同代数成人真皮成纤维细胞老化情况,采用Westernblot检测其AQP1表达情况,利用流式细胞术检测细胞荧光强度以观察细胞膜通透性变化。结果:与空白血清对照组同代细胞比较,二至丸含药血清组衰老细胞阳性细胞率降低(P<0.01),AQP1表达升高,细胞膜通透性下降(P<0.01)。结论:二至丸含药血清可延缓成人真皮成纤维细胞衰老,可能与其上调AQP1表达,降低细胞膜通透性,从而调节细胞内水平衡有关。Objective:To explore the effects of Erzhi Pill serum on aquaporin 1(AQP1)and cell membrane permeability in natural aging cell model.Methods:Erzhi Pill serum intervened adult dermal fibroblasts.The aging of adult dermal fibroblasts in different generations were observed.The expression of AQP1 was detected by Western blot.Flow cytometry was used to detect cell fluorescence intensity to observe the change of cell membrane permeability.Results:Compared with the same generation cells in the blank drug serum control group,the positive rate of aging cells in Erzhi Pill serum group was decreased(P<0.01),the expression of AQP1 was increased,and the cell membrane permeability was decreased(P<0.01).Conclusion:Erzhi Pill serum can delay adult dermal fibroblasts aging,which may be related to up-regulating AQPl expression and reducing cell membrane permeability,result in adjusting intracellular water balance.
