摘 要:目的:研究Th17细胞和Treg细胞在帕金森病模型小鼠外周血的表达状况,并从Th17/Treg平衡角度探讨针灸治疗帕金森病的潜在机制。方法:8周龄SPF级雄性C57BL/6小鼠随机分为空白组、模型组、针刺组、针灸组和雷沙吉兰组。采用MPTP腹腔注射的方法建立帕金森病动物模型,模型组不做治疗,仅作抓取固定;针刺组针刺百会、双侧阳陵泉穴;针灸组在针刺组基础上艾灸关元穴;雷沙吉兰组予雷沙吉兰溶液灌胃。观测各组小鼠疲劳转棒实验评分(ORP),采用流式细胞术检测各组小鼠外周血CD4^(+)CD25^(+)Foxp3^(+)Treg、CD4^(+)RORγt^(+)Th17细胞数,采用ELISA法检测外周血肿瘤坏死因子α(TNF-α)、白介素(IL)-17、IL-22、IL-4、γ干扰素(IFN-γ)、IL-10、转化生长因子β(TGF-β)的表达情况。结果:模型组小鼠ORP评分显著低于空白组(P<0.05),针刺组和针灸组小鼠ORP评分显著高于模型组(P<0.05)。与空白组比较,模型组小鼠外周血CD4^(+)CD25^(+)Foxp3^(+)Treg细胞数量显著减少(P<0.05),CD4^(+)RORγt^(+)Th17细胞数量显著升高(P<0.05);与模型组比较,雷沙吉兰组、针刺组、针灸组CD4^(+)CD25^(+)Foxp3^(+)Treg细胞数量均显著增加,CD4^(+)RORγt^(+)Th17细胞数量显著减少(P<0.05)。与正常组比较,模型组外周血TNF-α、IL-17、IL-22、IL-4、IFN-γ表达均显著升高(P<0.05),IL-10、TCF-β均显著降低(P<0.05)。与模型组比较,针刺组、针灸组、雷沙吉兰组外周血TNF-α、IL-17、IL-22、IL-4、IFN-γ表达均显著降低(P<0.05),IL-10、TGF-β均显著升高(P<0.05)。结论:针灸能够改善MPTP诱导的PD模型小鼠运动表现,并可调节Th17和Treg细胞分化和相关细胞因子的表达。Objective:To investigate the expression status of Th17 cells and Treg cells in the peripheral blood of mice with Parkinson's disease model and to explore the potential mechanism of acupuncture for the treatment of Parkinson's disease from the perspective of Th17/Treg homeostasis.Methods:Eight-week-old SPF-grade male C57BL/6 mice were randomly divided into blank group,model group,acupuncture group,acupuncture and moxibustion group and resagiline group.The animal model of Parkinson's disease was established by intraperitoneal injection of MPTP,and the model group was not treated but only fixed by grasping;the acupuncture group was treated with acupuncture points Baihui(GV 20)and both sides'Yanglingquan(GB 34);the acupuncture and moxibustion group was treated with moxibustion points Guan Yuan on the basis of the acupuncture group;the resagiline group was treated with resagiline solution by gavage.The ORP scores of mice in each group were observed,and the number of CD4^(+)CD25^(+)Foxp3^(+)Treg and CD4^(+)RORγt^(+)Th17 cells in peripheral blood of mice in each group was detected by flow cytometry.The expression of cytokines such as TNF-α,IL-4,IL-17,IL-22,IL-10,TGF-βand IFN-γ was measured by ELISA.Results:The ORP scores of mice in the model group were significantly lower than those in the blank group(P<0.05),and the ORP scores of mice in the acupuncture and acupuncture moxibustion groups were significantly higher than those in the model group(P<0.05).Compared with the blank group,the number of CD4^(+)CD25^(+)Foxp3^(+)lymphocytes in the peripheral blood of mice in the model group was significantly reduced(P<0.05),and the number of CD4^(+)RORγt^(+)lymphocytes was significantly increased(P<0.05);The number of CD4^(+)CD25^(+)Foxp3^(+)lymphocytes was significantly increased in the resagiline,acupuncture and acupuncture moxibustion groups compared with the model group,the number of CD4^(+)RORγt^(+)lymphocytes was significantly reduced(P<0.05).Compared with the blank group,peripheral blood TNF-α,IL-4,IL-17,IL-22 and IF
