摘 要:目的:观察前交叉韧带切断(ACLT)模型小鼠不同阶段关节软骨的病理变化特征,明确温经通络汤对小鼠膝骨关节炎的治疗作用。方法:将小鼠随机分为空白组、假手术组(术后1、2、4、8、12周)、模型组(术后1、2、4、8、12周)和中药低、高剂量组。ACLT法建立模型,化学染色观察关节软骨病理形态并采用改良Mankin’s评分评价,免疫组化分析COLⅡ的表达;Western blot及qPCR检测COLⅡ、COLⅩ、INOS、COX-2蛋白及mRNA的表达。结果:模型组小鼠软骨破坏程度较空白组随时间延长不断加深,COLⅡ表达随病程发展不断下降(P<0.01,P<0.05),COLⅩ水平持续升高(P<0.01);中药干预后软骨退变程度较模型组均有好转,中药高剂量组COLⅡ表达显著升高(P<0.01,P<0.05),COLⅩ、INOS、COX-2上调均被抑制(P<0.01,P<0.05)。结论:在ACLT模型小鼠中,关节病变呈时间依赖性加深,温经通络汤可通过改善关节炎症、延缓软骨退变干预膝骨关节炎进程。Objective: To observe the pathological changes of articular cartilage at different stages in anterior cruciate ligament transection(ACLT) mice model, and to determine the therapeutic effects of Wenjing Tongluo Decoction on knee osteoarthritis in mice. Methods: The mice were randomly divided into blank group, sham-operated group(1, 2, 4, 8 and 12 weeks after surgery), model group(1, 2, 4, 8 and 12 weeks after surgery) and low and high dose groups of Chinese medicine. The models were established by ACLT method. The pathological morphology of articular cartilage was observed by chemical staining and evaluated by modified Mankin’s score. The expression of COLⅡ was analyzed by immunohistochemistry. The protein and expressions of COLⅡ, COLⅩ, INOS and COX-2 were determined by Western blot and qPCR. Results: The degree of cartilage destruction in the model mice continued to deepen with time than that in the blank group, and the expression of COLⅡ decreased continuously with the development of the disease(P<0.01, P<0.05), while the levels of COLⅩ increased with time(P<0.01). After TCM intervention, the degree of cartilage degeneration was improved compared with the model group, the expression of COLⅡ was back increased(P<0.01, P<0.05) and the upregulation of COLⅩ, INOS and COX-2 were all inhibited(P<0.01, P<0.05) in low and high dose groups. Conclusion: In ACLT model mice, the joint lesions deepened in a time-dependent manner, and Wenjing Tongluo Decoction can interfere with the progression of knee OA by ameliorating joint inflammation and delaying cartilage degeneration.
