摘 要:目的:探讨脑泰方(NTF)抑制局灶性脑缺血后炎症因子表达机制。方法:将SD大鼠随机分为假手术组、模型组、NTF低剂量组(3g/kg)、NTF中剂量组(9g/kg)、NTF高剂量组(27g/kg)。各组大鼠预处理灌胃给药连续3d,大脑中动脉闭塞(MCAO)模型制备术后灌胃给药1d,取材。HE染色检测病理组织形态;免疫组织化学染色检测各组大脑皮质信号转导和转录激活因子3磷酸化(p-STAT3)、白介素-6(IL-6)蛋白的表达;Western blot检测p-STAT3、IL-6、肿瘤坏死因子α(TNF-α)表达。结果:NTF中、高剂量组中皮质缺血区椎体细胞胞体皱缩和核固缩较模型组少。与模型组比较,NTF中、高剂量组大鼠大脑皮质中IL-6和p-STAT3均表达水平显著下调(P<0.05);NTF中、高剂量组p-STAT3蛋白表达水平显著降低(P<0.05,P<0.01),NTF高剂量组IL-6蛋白表达水平显著下降(P<0.01),NTF中、高剂量组TNF-α蛋白表达水平显著降低(P<0.01)。结论:脑泰方可以通过抑制STAT3蛋白磷酸化降低脑缺血后炎症反应,起到神经元保护作用。Objective:To explore the mechanism of Naotaifang(NTF)inhibiting the expression of inflammatory factors after focal cerebral ischemia.Methods:SD rats were randomly divided into normal group,model group,low dose of NTF group(3 g/kg),medium dose of NTF group(9 g/kg)and high dose of NTF group(27 g/kg).Each group was given intragastric administration 3 days before operation,after the operation of middle cerebral artery occlusion(MCAO),intragastric administration was given 1 day,and specimens of cerebral cortex were harvested since last intragastric administration.HE staining was used to detect the pathological morphology.The expression of p-STAT3 and IL-6 protein in cerebral cortex of each group was detected by immunohistochemical staining.The protein expression levels of p-STAT3,IL-6 and TNF-α were detected by Western blot.Results:There were fewer shrinkage and nuclear pyopysis of vertebral cells in the cortical ischemia area in medium and high dose NTF groups than that in the model group.Compared with model group,the expression levels of IL-6 and p-STAT3 were down-regulated in medium and high dose NTF groups(P<0.05),the protein expression level of p-STAT3 in medium and high dose NTF groups was decreased(P<0.05,P<0.01),the protein expression level of IL-6 in high dose NTF group was significantly decreased(P<0.01),and the protein expression level of TNF-α in medium and high dose NTF groups was significantly decreased(P<0.01).Conclusion:NTF can ameliorate inflammation to protect neuron of cerebral cortex in cerebral ischemia,which may be related to the inhibition phosphorylationof STAT3.
