摘  要:采用分光光度法,以齐墩果酸为对照物,香草醛-冰醋酸-高氯酸为显色体系,测定青叶胆中总三萜的含量。在单因素试验的基础上以料液比、提取时间、乙醇浓度为考察因素,总三萜酸提取率为评价指标,采用Box-Behnken响应面法设计试验方法,采用minitab软件对数据进行处理,建立二次回归模型。结果显示二次回归模型显著,预测性较强,总三萜酸的最佳提取工艺为料液比14mL/g、提取时间66min、乙醇浓度60%。结论:利用响应面法优选出的提取工艺简单、合理,增加了提取率。The content of total triterpenoids was determined by UV with oleanolic acid as reference. Vanilla aldehyde-glacial acetic acid-perchloric acid for color development system. On the basis of single factor, ratio of material to liquid, extraction time, ethanol concentration as the research factors. Yield of total triterpenoids was used as the evaluation index. Box-behnken response surface method was used to design the test method, minitab software was used to process the data, and establish the quadratic regression model. The quadratic regression model is remarkable and has strong predictability. The optimum process: ratio of liquid14mL/g, extraction time of 66min, ethanol concentration of 60%. Conclusion: The extraction process optimized by response surface methodology is simple and reasonable, increased extraction rate.
