摘 要:为了解凡纳滨对虾感染白斑综合症病毒(white spot syndrome virus,WSSV)后病毒的增殖变化,采用饲喂法对凡纳滨对虾进行WSSV人工感染,采用real-time PCR方法检测感染初期、高峰期和后期死亡对虾体内的病毒含量.结果表明,随着感染时间的延长,感染初期、高峰期和后期对虾体内WSSV含量范围分别为1.37×105~4.47×10^(6)、1.92×10^(6)~1.30×10^(7)和3.75×10^(6)~1.90×10^(7)copies·ng^(-1),平均病毒含量分别为1.56×10^(6)、7.56×10^(6)和1.02×10^(7)copies·ng^(-1),感染后期对虾体内的WSSV含量显著高于感染初期(P=0.023),且感染后期死亡的对虾对WSSV的耐受力更强.In order to understand the changes in the proliferation of white spot syndrome virus(WSSV)after infection with White spot syndrome virus(WSSV)in Litopenaeus vannamei,a total of 487 shrimp individuals(1.5-2.8 g)of L.vannamei were artificially infected with WSSV.According to the shrimp death curve,shrimp were divided into three phages:early,peak and late phage.The WSSV load in 20 samples of three infection stages were quantified by absolute real-time PCR.The WSSV loads in each stages were 1.37×10^(5)-4.47×10^(6)、1.92×10^(6)-1.30×10^(7)、3.75×10^(6)-1.90×10^(7)copies·g^(-1)DNA,with the average WSSV loads of 1.56×10^(6)、7.56×10^(6)、1.02×10^(7)copies·g^(-1)DNA,respectively.The WSSV loads in late stage were significantly higher than that in early stage(P=0.023).The results also showed that shrimp in late stage had better WSSV resistance.This study will be helpful to reveal the occurrence of WSSV and provide the theoretical value for WSSV resistant breeding of L.vannamei.
