摘 要:为建立细基江蓠繁枝变种遗传转化体系,以细基江蓠繁枝变种类愈伤组织为受体材料,采用农杆菌介导的方法,建立一种细基江蓠繁枝变种遗传转化方法.研究结果发现,往细基江蓠繁枝变种培养液中添加20μmol·L^(-1)茉莉酸甲酯、2.0 mg·L^(-1)2,4二氯苯氧乙酸、0.5 mg·L^(-1)激动素的植物激素组合,其类愈伤组织诱导效果最好,诱导率高达56.98%.类愈伤组织转化子的GFP绿色荧光检测和PCR鉴定结果均为阳性.这说明细基江蓠繁枝变种外植体的类愈伤组织可作为受体材料,采用农杆菌介导方法可实现外源gfp基因在类愈伤组织中的表达,为江蓠品种的遗传改良提供一个强有力的工具.In order to establish the genetic transformation system of Gracilaria tenuistipitata var.liui Zhang et Xia,the callus like tissues induced from the explants of Gracilaria tenuistipitata were used as receptor materials,and an agrobacterium mediated genetic transformation method of Gracilaria tenuistipitata was established The study found that the plant hormone combination of 20μmol·L^(-1)methyl jasmonate,2.0 mg·L^(-1)2,4 dichlorophenoxyacetic acid and 0.5 mg·L^(-1)kinetin added to the culture medium had the best callus like induction effect,with the induction rate as high as 56.98%.GFP green fluorescence detection and PCR identification of calli like transformants were positive.The experimental results showed that the callus like tissues induced from the explants of Gracilaria tenuistipitata could be used as receptor material,and the expression of exogenous gfp gene in callus could be realized by Agrobacterium mediated transformation method,which provided a powerful tool for the genetic improvement of Gracilaria.
