摘 要:抗生素抗性基因(ARGs)污染可对人类健康造成潜在威胁,分析了水处理过程中加入的杀菌剂戊二醛对环境微生物及ARGs分布的影响,加入戊二醛会使混菌体系的微生物数量降低,并将ARGs相对浓度提升2个数量级,意味着单位菌体内含有更多的ARGs.而添加纳米零价铁(nZVI),能够有效抑制戊二醛处理后的混菌体系ARGs浓度上升现象,ARGs相对浓度仅是加入戊二醛处理后的22.5%.流式细胞术和转录组分析结果表明,0.1%戊二醛的加入使分离出的铜绿假单胞菌DQ14以细胞膜损伤的状态存在,编码外排泵组成蛋白的tolC基因显著上调了6.2倍.研究表明nZVI可有效降低油田水处理系统中使用戊二醛造成的ARGs传播风险.Antibiotic resistance genes(ARGs)pollution may be a major threat to human health.The effects of adding glutaraldehyde as bactericide in the water treatment process on bacteria and ARGs were analyzed.The addition of glutaraldehyde led to a sharp decrease in the number of bacteria and an increase of two orders of magnitude in the relative concentration of ARGs,which indicated that each bacteria contained more ARGs.The addition of nano zero valent iron(nZVI)could inhibit the increase of ARGs concentration after glutaraldehyde treatment.When adding nZVI together with glutaraldehyde,the relative concentration of ARGs was 22.5%of that of adding glutaraldehyde only.Results of flow cytometry and transcriptome analysis revealed that the additionof glutaraldehyde led to damage of Pseudomonas aeruginosa DQ14's cell membrane,also known as causing intermediate state.What's more,the tolC gene encoding the efflux pump component protein was significantly up-regulated by 6.2 times.The research shows that NZVI can effectively reduce the risk of ARGs transmission caused by glutaraldehyde in oilfield water treatment system.
