摘  要:目的探究清脑益智方通过调控乙酰胆碱等相关胆碱能物质,改善阿尔茨海默症的作用机制。方法以SAMR1小鼠为对照,将SAMP8小鼠分为高剂量清脑益智方组、低剂量清脑益智方组、模型组,共4组,每组各10只,连续灌胃1个月后,行Mirros水迷宫检测小鼠学习记忆等能力变化,免疫组化法检测Aβ沉积情况,酶联免疫吸附法检测乙酰胆碱(Ach)含量、比色分析法检测乙酰胆碱酯酶(AchE)、乙酰胆碱转移酶(ChAT)活力,蛋白印迹法检测突触素(SYN)、生长相关蛋白-43(GAP-43)的表达。结果与模型组相比,经清脑益智方干预后,(1)高剂量组小鼠停留平台所在象限时间延长,穿越平台次数增加,潜伏期缩短(P<0.05);(2)不同剂量组小鼠脑内Ach含量均有所提高,低剂量组效果更明显(P<0.05);(3)不同剂量组小鼠脑内AchE活力均有降低趋势(P>0.05);(4)不同剂量组小鼠脑内ChAT活力均提高(P<0.05);(5)不同剂量组小鼠脑内SYN含量均升高,且高剂量效果更明显(P<0.05);(6)不同剂量组小鼠脑内GAP-43的含量均提高(P<0.05)。结论清脑益智方改善阿尔茨海默症的学习记忆能力,提高SYN、GAP-43的表达,提高胆碱能系统ChAT的表达,增加Ach的合成、释放,其机制与调节突触可塑性,改善胆碱能神经元及神经递质功能有关。Objective To explore the mechanism of Qingnao Yizhi Formula(清脑益智方)regulating acetylcholine and other related cholinergic substances to improve Alzheimer′s disease.Methods Taking SAMR1 mice as a control,SAMP8 mice were divided into Qingnao Yizhi Formula high-dose group,Qingnao Yizhi Formula low-dose group,and model group,a total of 4 groups,with 10 rats in each group.And 1 continuous intragastric administration month later,the Mirros water maze was used to detect the changes in learning and memory of mice.The immunohistochemical method was used to detect Aβdeposition.The enzyme-linked immunosorbent assay was used to detect the acetylcholine content and the colorimetric analysis to detect the activities of acetylcholinesterase and acetylcholine transferase.Western blotting was used to detect the expressions of synaptophysin and growth-associated protein-43.Results Compared with the model group,after the intervention of Qingnao Yizhi Formula,(1)the mice in the high-dose group stayed longer in the quadrant of the platform.The times of crossing the platform and the incubation period were shortened(P<0.05).(2)The content of Ach was increased,and the effect was more obvious in the low-dose group(P<0.05).(3)The activity of acetylcholinesterase(AchE)in the brain of mice in different dose groups decreased(P>0.05).(4)The activities of choline acetyltransferase(ChAT)in the brain of mice in different dose groups were all increased(P<0.05).(5)The content of SYN in the brain of mice in different dose groups increased,and the effect of high-dose group was more obvious(P<0.05).(6)The contents of growth associated protein 43(GAP-43)in the brain of mice in different dose groups were increased(P<0.05).Conclusion Qingnao Yizhi Formula can improve the learning and memory ability of Alzheimer′s disease,increase the expressions of SYN and GAP-43,increase the expression of ChAT in the cholinergic system,and increase the synthesis and release of Ach.Its mechanism is related to regulating synaptic plasticity and improving th
