摘 要:目的通过观察增率复脉颗粒对病态窦房结综合征兔模型心肌细胞钙离子通道Cav1.2、Cav1.3和Cav3.1、Cav3.2、Cav3.3蛋白及mRNA表达的影响来探讨其治疗病态窦房结综合征的机制。方法选择60只雄性大耳白家兔,用40%甲醛液外敷窦房结区表面造成病态窦综合征模型,随机分为正常组、模型组、阳性药组(心宝丸)、增率复脉颗粒中、低剂量组(2.66 g/kg、1.33 g/kg),造模后检测心率,验证是否造模成功。造模后稳定10 min后给药,观察给药后30、60、90、120 min心率,并取标本,采用Western blot法及实时荧光定量聚合酶链式反应(qPCR)观察5组兔模型给药后心肌细胞Cav1.2、Cav1.3和Cav3.1、Cav3.2、Cav3.3蛋白及mRNA表达的变化。结果与模型组比较,阳性药(心宝丸)组、增率复脉颗粒中、低剂量组在给药后不同时间点均可提升心率,差异有统计学意义(P<0.05)。与模型组比较,各给药组钙离子通道Cav1.2 mRNA表达均升高,差异有统计学意义(P<0.05);阳性药(心宝丸)组钙离子通道Cav1.3、Cav3.1、Cav3.2 mRNA表达均升高,差异有统计学意义(P<0.05);钙离子通道Cav3.3 mRNA表达无明显变化,差异无统计学意义(P>0.05)。与模型组比较,Western-blot法检测阳性药(心宝丸)组心肌细胞钙离子通道Cav1.2、Cav3.1蛋白表达均升高,差异有统计学意义(P<0.05);阳性药(心宝丸)组、增率复脉颗粒中剂量组钙离子通道Cav1.3,3.2蛋白表达均升高,差异有统计学意义(P<0.05);钙离子通道Cav3.3蛋白表达无明显变化,差异无统计学意义(P>0.05)。结论中药(心宝丸、增率复脉颗粒)可提升心率,调控心肌细胞钙离子通道可能是中药治疗病态窦房结综合征的机制之一。Objective To observe the effect of Zenglv Fumai Granules(ZLFM)on the expression of Cav1.2,Cav1.3,Cav3.1,Cav3.2,and Cav3.3 proteins and mRNAs in myocardial cells of rabbits with sick sinus syndrome(SSS),and to study its mechanism in the treatment of the disease.Methods Sixty male rabbits were randomly divided into normal group,model group,positive drug group,ZLFM medium-dose groups(2.66 g/kg)and low-dose groups(1.33 g/kg).Except the normal group,rabbits in the other groups were treated by external application of 40%for malin to the surface of sinoatrial node to induce SSS model.After modeling,the heart rate of rabbits was measured to verify the modeling.Ten minutes after successful modeling,the SSS model rabbits were administered with drugs,and the heart rate was measured at 30,60,90,and 120 minutes after administration.The expression of Cav1.2,Cav1.3,Cav3.1,Cav3.2 and Cav3.3 proteins and mRNAsin the five groups was observed by Western blot and real-time fluorescence quantitative polymerase chain reaction(real-time qPCR),respectively.Results Compared with model group,positive drug(Xinbao Pills)group,ZLFM medium-dose groups and low-dose groups had increased heart rate at different time points after administration(P<0.05).Compared with the conditions in model group,the expression of Cav1.2 mRNA in administration groups was increased(P<0.05),and the expression of Cav1.3,Cav3.1 and Cav3.2 mRNAs in positive drug group was also increased(P<0.05),while there was no significant difference in the expression of Cav3.3 mRNA(P>0.05).Western-blot revealedthat the expression of Cav1.2 and Cav3.1 proteins in positive drug group was higher(P<0.05)and the expression of Cav1.3 and Cav3.2 proteins in positive drug group and ZLFM medium-dose group was also higher than those in model group(P<0.05),while no marked difference was observed in the expression of Cav3.3 protein(P>0.05).Conclusion Chinese medicines(Xinbao Pills,ZLFM Granules)elevated the heart rate of SSS rabbits,and their mechanism might be regulating the calcium channels i
