摘 要:目的 观察益气化瘀解毒方对脂多糖(lipopolysaccharide, LPS)诱导的急性呼吸窘迫综合征(acute respiratory distress syndrome, ARDS)模型大鼠的保护作用及对窖蛋白-1(caveolin-1, Cav-1)、白细胞介素-12(interleukin-12, IL-12)、白细胞介素-13(interleukin-13, IL-13)表达水平的影响并探讨其作用机制。方法 18只SD大鼠随机分为对照组、模型组及中药复方组3组,每组6只。中药复方组予益气化瘀解毒方12.2 g/(kg·d)预防性灌胃给药,其余组等量蒸馏水灌胃。尾静脉注射LPS建立ARDS模型,造模后16小时处死大鼠收集标本,光镜下观察肺组织病理学改变,采用酶联免疫法检测肺泡灌洗液、肺匀浆Cav-1及血清IL-12、IL-13表达水平,采用免疫印迹法检测肺匀浆Cav-1蛋白表达水平。结果 益气化瘀解毒方可减少ARDS模型大鼠肺泡结构破坏、肺水肿及炎性细胞浸润,且与模型组相比,中药复方组Cav-1蛋白表达显著上调(P<0.05),促炎因子IL-12表达显著减少(P<0.01);与对照组相比,抑炎因子IL-13表达增加(P<0.01)。结论 益气化瘀解毒方对LPS诱导的ARDS模型大鼠具有保护作用,可能与调节Cav-1蛋白及炎症因子IL-12、IL-13的表达水平有关。Objective To observe the protective effects of YiqiHuayuJiedu Decoction(YQHYJD) on LPS-induced ARDS rats and the expression levels of Cav-1, IL-12 and IL-13.Methods 18 male SD rats were randomly divided into 3 groups: controlled group, model group and TCM formula group. The rats in the TCM formula group was administrated with YQHYJD once every day, while corresponding distilled water were given respectively in other groups. The ARDS rat model was established by tail vein injection of LPS. The pathological changes of the lung tissues were observed under light microscope, and the expression levels of Cav-1 in bronchoalveolar lavage fluid(BALF) and lung homogenate as well as IL-12 and IL-13 in serum were detected by the method of ELISA, while the expression levels of Cav-1 in lung homogenate were also detected by Western Blotting.Results The results of HE staining showed that YQHYJD could reduce pulmonary alveolar damage, pulmonary edema and inflammatory cells infiltration. Compared with the model group, the protein expression levels of Cav-1 were increased(P<0.05) and the expression levels of pro-inflammatory factors IL-12 were decreased(P<0.01), while compared with the controlled group, the expression levels of anti-inflammatory factors IL-13 were increased(P<0.01) in TCM formula group.Conclusion YQHYJD has protective effects on LPS-induced ARDS rats, which may be related to the regulation of the expression levels of Cav-1, IL-12 and IL-13.
