摘 要:目的分析高迁移率族蛋白B1(HMGB1)单抗对大鼠脑出血术后脑水肿的作用。方法将40只健康雄性成年SD大鼠按照随机数字表法分为实验组和对照组,各20只。所有大鼠采用改良Miller法建立脑出血术后脑水肿模型,而后实验组大鼠采用HMGB1单抗处理,对照组大鼠仅给予等体积的0.9%氯化钠溶液处理。对比2组大鼠术后72、120、168 h的脑水肿体积,血清HMGB1、白介素6(IL-6)、肿瘤坏死因子α(TNF-α)水平及脑组织水通道蛋白4(AQP-4)、基质金属蛋白酶9(MMP-9)表达水平。结果术后120、168 h,实验组大鼠脑水肿体积小于对照组,血清HMGB1、IL-6、TNF-α水平低于对照组(P<0.01)。术后72、120、168 h,实验组大鼠脑组织AQP-4、MMP-9表达水平低于对照组(P<0.01)。结论HMGB1单抗可有效控制脑出血术后大鼠脑水肿,抑制机体炎性反应,调控AQP-4、MMP-9表达,进而减轻脑损伤。Objective To analyze the effect of HMGB1 monoclonal antibody on brain edema after intracerebral hemorrhage in rats.Methods A total of 40 healthy male adult SD rats were randomly divided into experimental group and control group,20 rats in each group.All rats were established cerebral hematoma model after intracerebral hemorrhage by modified Miller′s method,and then rats in experimental group were given HMGB1 monoclonal antibody,and rats in control group were given equal volume of 0.9%sodium chloride solution.Brain edema volume,serum levels of HMGB1,IL-6,TNF-α,expression levels of AQP-4,MMP-9 in brain tissue at 72,120,168 h after operation were compared between the two groups.Results At 120,168 h after operation,brain edema volume in the experimental group was less than that in the control group,serum levels of HMGB1,IL-6,TNF-αwere lower than those in the control group(P<0.01).At 72,120,168 h after operation,expression levels of AQP-4,MMP-9 in brain tissue in the experimental group were lower than those in the control group(P<0.01).Conclusion HMGB1 monoclonal antibody can effectively control brain edema after cerebral hemorrhage,inhibit inflammatory reaction,regulate the expression of AQP-4 and MMP-9,and then reduce brain injury.
