摘 要:目的探讨烧伤后增生性瘢痕(HS)组织中颗粒蛋白前体(PGRN)、环氧合酶2(COX-2)的表达水平及其与炎性因子的相关性。方法收集54例烧伤后HS患者的HS组织与正常皮肤组织。采用免疫组织化学法检测各组织中的PGRN、COX-2蛋白阳性表达率,采用实时荧光定量PCR检测各组织中的PGRN、COX-2、白细胞介素(IL)-6、IL-10、IL-1β和肿瘤坏死因子α(TNF-α)的mRNA表达水平。分析HS组织中PGRN、COX-2、IL-6、IL-10、IL-1β、TNF-α的mRNA表达水平的相关性。结果HS组织中的PGRN、COX-2蛋白阳性表达率,以及PGRN、COX-2、IL-6、IL-1β、TNF-α的mRNA表达水平均高于正常皮肤组织,IL-10 mRNA表达水平低于正常皮肤组织(均P<0.05)。HS组织中的PGRN mRNA表达水平与COX-2 mRNA表达水平呈正相关,且PGRN、COX-2的mRNA表达水平与IL-6、IL-1β、TNF-α的mRNA表达水平均呈正相关,与IL-10 mRNA表达水平均呈负相关(均P<0.05)。结论PGRN和COX-2在HS组织中的表达上调,两者可能通过调节IL-1β、IL-6、TNF-α等炎性因子的产生,促进炎症反应,共同参与烧伤后HS的形成。Objective To investigate the expressions of progranulin(PGRN)and cyclooxygenase 2(COX-2)in hypertrophic scar(HS)tissues after burn and their correlation with inflammatory factors.Methods The tissues of HS and normal skin of 54 HS patients after burn were collected.The immunohistochemistry was used to detect the positive expression rates of PGRN and COX-2 proteins in various tissues.The real-time fluorescent quantitative PCR was employed to detect the mRNA expressions of PGRN,COX-2,interleukin(IL)-6,IL-10,IL-1βand tumor necrosis factorα(TNF-α)in various tissues.The correlation between the mRNA expressions of PGRN,COX-2,IL-6,IL-10,IL-1βand TNF-αin HS tissues was analyzed.Results The positive expression rates of PGRN and COX-2 proteins,as well as the mRNA expressions of PGRN,COX-2,IL-6,IL-1β,and TNF-αin HS tissues were all higher than those in normal skin tissues,whereas the mRNA expression of IL-10 was lower in HS tissues than in normal skin tissues(all P<0.05).The mRNA expression of PGRN positively correlated with the mRNA expression of COX-2 in HS tissues,and the mRNA expressions of PGRN and COX-2 positively correlated with the mRNA expressions of IL-6,IL-1βand TNF-α,whereas negatively correlated with the mRNA expression of IL-10(all P<0.05).Conclusion PGRN and COX-2 are up-regulated in HS tissues,and the two may promote inflammatory responses and be involved in the formation of HS after burn by regulating the production of inflammatory factors as for IL-1β,IL-6,and TNF-α,etc.
