摘 要:WOX转录因子作为植物特异性转录因子,在植物生长发育及非生物胁迫响应过程中发挥了重要作用,探究珍珠粟WOX家族基因的详细特征及功能有助于进一步解析其在珍珠粟生长发育各个环节中的调控过程.本研究在珍珠粟基因组中共鉴定得到15个WOX家族基因成员.利用生物信息学方法对蛋白理化性质、亚细胞定位分析表明,珍珠粟WOX家族蛋白成员大部分为不稳定酸性亲水蛋白,大多定位在细胞核中.对基因上游启动子区域分析表明,在WOX家族基因启动子区域存在大量与光响应、植物激素响应、抗逆性、生长发育相关的顺式作用元件.保守基序分析发现了2个构成WOX家族基因成员特有结构域的基序.为探究珍珠粟与其他物种的进化关系,构建珍珠粟、拟南芥、水稻、紫象草物种间系统发育进化树并将其分为古代支、中间支、现代支,同一分支下基因成员基序的个数、次序具有相似性.实时荧光定量PCR检测表明,珍珠粟WOX家族基因成员的表达模式具有明显的组织特异性.本研究为深入探讨珍珠粟WOX家族基因的功能提供了有价值的信息.As plant-specific transcription factor,WUSCHEL related homeobox(WOX)play an important role in plant growth,development and response to abiotic stresses.To elucidate its role in the growth and development of Cenchrus americanus,the WOX gene family members were genome-wide identified in C.americanus for bioinformatics analysis of their protein physicochemical properties and subcellular localization.The results showed that the majority of the 15 identified WOX proteins in C.americanus were unstable acidic hydrophilic and localized in the nucleus.And the promoter region of these WOX genes harbored a large number of cis-acting elements related to photoresponse,plant hormones,stress resistance and growth.Two conserved motifs were identified in the structural domain which was specific to the WOX family.Phylogenetic analysis based on WOX genes in C.americanus,Arabidopsis thaliana,Oryza sativa and Pennisetum purpureum cv.‘Purple’showed 3 major clades of ancient,medial and modern,and the number,position and order of motifs were similar between the species under the same clade.Furthermore,quantitative real-time PCR analysis revealed the tissue specific expression patterns of WOX,indicating the diverse biological functions of WOX in C.americanus.This study provided valuable information for an in-depth study of the functions of the WOX family genes in C.americanus.
