摘 要:为探究缺乏雌激素对雄性中华鳖脂代谢的影响,通过腹腔注射不同剂量的芳香化酶抑制剂和雌激素受体拮抗剂,检测芳香化酶基因(Cyp19a1基因)、脂代谢相关基因的表达差异及肝脏组织学差异。组织切片染色结果显示,雌激素受体拮抗剂处理后,肝脏着色加深,表明肝脏产生脂肪堆积,且呈剂量效应。实时荧光定量PCR结果显示,雌激素受体拮抗剂处理1 d后,随着剂量的增加,Cyp19a1基因的表达量显著降低(P<0.05),但4 d后和42 d后显著升高(P<0.05);脂肪酸合成酶基因(Fas基因)、脂肪沉积相关基因(Scd和Pparγ基因)与脂肪分解相关基因(Stat3基因)表达量在雌激素受体拮抗剂处理42 d后显著升高(P<0.05)。低剂量芳香化酶抑制剂处理49 d后,Cyp19a1基因表达量显著升高(P<0.05),高剂量组表达量显著降低(P<0.05);Fas基因随剂量增加表达量呈升高趋势,处理后49 d高剂量组表达量降低;处理后49 d,处理组Pparγ基因和Bmp4基因的表达量呈降低趋势,高剂量组Scd基因表达量升高。定量结果表明,雌激素代谢紊乱会导致脂代谢失调,并与雌激素剂量有关。本试验结果可为中华鳖体内雌激素和脂代谢之间的关系研究提供理论参考。We investigated the effects of estrogen deficiency on lipid metabolism in Chinese soft-shelled turtle Pelodiscus sinensis by intraperitoneal injection of different concentrations of Letrozole(LET)and Fulvestrant(ICI)to detect differences in the expression of Cyp19a1 genes related to lipid metabolism and liver histology.Liver staining deepened in stained histological sections after ICI treatment,indicating that fat accumulation was produced in the liver and that this accumulation was dose-dependent.RT-qPCR results showed that Cyp19a1 expression level was significantly lower with the increase in treatment concentration(P<0.05)at 1 d after ICI treatment,significantly higher(P<0.05)at 3 d and 42 d.Similarly,as the ICI treatment concentration increased,the expression levels of Fas,Scd,Pparγand Stat3 genes were significantly higher(P<0.05).Cyp19a1 expression level was significantly higher(P<0.05)after 42 d of treatment with low concentrations of LET and significantly lower(P<0.05)with high concentrations of LET.The expression level of Fas gene was increased with the increase in treatment concentration and was significantly lower in the 49 d in high concentration group(P<0.05).In the 49 d,the expression levels of Scd and Pparγwere decreased in treatment concentration group,and the expression level of Stat 3 was increased in high concentration group.RT-qPCR results revealed that disturbances in estrogen metabolism led to dysregulation of lipid metabolism and were associated with estrogen concentrations.The findings provided a theoretical reference for discussing the relationship between estrogen and lipid metabolism and breeding in the soft-shelled turtle.
