摘 要:YABBY基因是植物所特有的一类转录因子家族,在植物器官形成、生长发育、信号转导以及胁迫响应等生物学过程发挥着重要调控作用。野生二粒小麦(Triticum dicoccoides)作为普通小麦的四倍体祖先种,一直是小麦遗传改良的重要种质和基因库,但目前有关野生二粒小麦的YABBY基因还未见报道。为了挖掘野生二粒小麦的YABBY基因,本研究利用生物信息学方法对野生二粒小麦YABBY转录因子家族进行了全基因组鉴定和分析,结果在野生二粒小麦基因组中共鉴定到了12个YABBY基因,归属于6个同源基因对且位于5个同源染色体组上。系统进化分析将它们分成YAB1/3、YAB5和CRC 3个亚家族,而且同一亚家族成员间具有相似的结构特征。基于RNA-seq数据的表达特性分析发现,YABBY在野生二粒小麦不同组织中具有特异性表达;同时,还鉴定发现了3个YAB5亚族成员在盐胁迫下发生了差异表达,进一步的qPCR分析验证了它们的表达特性,表明它们可能是盐胁迫响应相关的YABBY转录因子。The YA BBY gene is a family of plant-specific transcription factor family has a prominent role in regu lating plant growth,organogenesis,signal transduction pathway and abiotic stress responses.Wild emmer(Triticum dicoccoides)as an ancestor of common wheat is an important germplasm for wheat genetic improvement.Howev-er,YABBY gene in wild emmer wheat has not been reported yet.In order to mine the YABBY gene in wild emmer wheat,YABBY transcription factor family was identified and analyzed in this study based on bioinformatics method.A total of 12 YA BBY(TdYABBY)genes were identified in wild emmer wheat,which were clustered into 6 orthologous gene pairs localizing on 5 homologous chromosomes.They could be divided into YAB1/3,YAB5 and CRC subfamilies through phylogenetic analysis and the members belonging into the same subfamily shared the similar gene structures.Furthermore,the expression pattern of these identified were investigated using RNA-seq data and they showed specific expression in different tissues.It is also found that 3 TdYA BBY genes in YAB5 sub-family displayed differential expression under salt stress treatment,and qPCR analysis verified their expression patterns,suggesting that they might be salt-responsive YABBY transcription factor in wild emmer wheat.
