摘 要:目的观察三七皂苷R1对血管平滑肌细胞(VSMC)迁移及p38 MAPK蛋白的影响。方法用AngⅡ诱导VSMC迁移建立细胞迁移模型,将迁移的VSMC随机分为对照组和三七皂苷R1低、中、高剂量组(5、10、20μmol/L)。用细胞划痕实验检测VSMC迁移面积,Western blotting法检测MMP2、MMP9、磷酸化p38 MAPK及p38 MAPK蛋白表达。结果与对照组相比,三七皂苷R1中、高剂量组均能明显减少VSMC的细胞迁移面积及MMP2、MMP9迁移蛋白表达(P<0.05),且呈剂量依赖性;与对照组相比,三七皂苷R1中、高剂量组均能明显减少p38 MAPK蛋白磷酸化(P<0.05),且呈剂量依赖性。结论三七皂苷R1抑制了AngⅡ诱导的VSMC迁移面积,同时下调MMP2和MMP9迁移蛋白,且呈剂量依赖性,其机制可能与降低p38 MAPK蛋白磷酸化有关。Objective:To investigate the effects of notoginsenoside R1(NGR1)on vascular smooth muscle cells(VSMCs)migration and p38 MAPK protein.Methods:VSMCs were induced by AngⅡto establish cells migration model.The migration VSMCs were randomly divided into the control group,NGR1 low-dose group(5μmo/L),medium-dose group(10μmo/L),and high-dose group(20μmo/L).The area of VSMCs migration were detected by cells scratch assay,and the MMP2,MMP9,phosphorylated p38 MAPK and p38 MAPK proteins were detected by Western blotting.Results:Compared with the control group,both NGR1 medium and high dose groups could significantly reduce the cell migration area and the MMP2 and MMP9 migration proteins(P<0.05),and the inhibitory effect were dose dependent.The phosphorylation of p38 MAPK protein was also significantly reduced(P<0.05)and dose dependent.Conclusion:NGR1 can inhibit AngⅡ-induced VSMC migration and reduce the MMP2 and MMP9 proteins to express in a dose-dependent manner,and the mechanism may be related to decrease the phosphorylation of p38 MAPK protein.
