摘  要:为探究桃儿七体细胞胚发生过程中内源激素的积累规律,提高体细胞胚诱导率,本试验采用超高效液相色谱-串联质谱(UPLC-MS/MS)法测定了桃儿七非胚性愈伤组织(NEC)、胚性愈伤组织(EC)、不完全致密胚性组织(ICpEC)和球形胚(GE)这4个阶段的生长素(IAA)及其类似物、细胞分裂素(BAP)及其类似物以及4种赤霉素(GA)和2种脱落酸(ABA)的含量,并探讨了4类内源激素在桃儿七体细胞胚发生过程中的调控作用,以期为桃儿七体细胞胚发生的激素调控提供理论依据。结果表明:生长素及其类似物促进了胚性细胞的形成,含量呈逐渐增加的趋势,高浓度的生长素促进了球形胚的形成;细胞分裂素及其类似物对体细胞胚的形成和保持具有关键作用,积累呈先增加后减小的趋势,积累量在胚性愈伤阶段最低,至不完全致密胚性组织阶段达到最高,球形胚阶段趋于稳定;赤霉素对桃儿七胚性细胞发生起反向调控作用,其中GA3在非胚性愈伤组织阶段积累量较高,在胚性愈伤组织阶段最低,较高浓度的赤霉素抑制体细胞胚的形成,在桃儿七体细胞胚发生的4个阶段呈先上升后下降的趋势;脱落酸含量在体细胞胚发生的4个阶段呈逐渐降低趋势,高浓度脱落酸促进了桃儿七体细胞胚的快速形成,而内源脱落酸浓度的逐渐降低使桃儿七愈伤组织的胚性得到维持,延缓了体细胞胚的成熟,这可能是导致畸形胚发生的原因之一。本研究结果为桃儿七体细胞胚的培养提供了理论依据。In order to investigate the accumulation pattern of endogenous hormones during somatic embryogenesis of Sinopodophyllum emodi(Wall.) Ying and improve the induction rate of somatic embryogenesis, contents of auxin(IAA) and its analogues, cytokinins(BAP) and its analogues, 4 kinds of gibberellins(GA), and 2 kinds of abscisic acid(ABA) in 4 stages of S. emodi namely non-embryonic callus(NEC), embryonic callus(EC), incomplete dense embryonic tissue(ICpEC), and spherical embryo(GE) were assayed by using an ultra-high liquid chromatography-tandem mass spectrometry(UPLC-MS/MS) method in this study. Meanwhile, the regulation effect of 4 classes of hormones on the somatic embryogenesis of S. emodi were discussed, which provided a theoretical basis for the hormone regulation of somatic embryogenesis of S. emodi. The results showed that auxin and its analogues promoted the formation of embryogenic cells, their contents showed a tendency to gradually increase, and high concentrations of auxin promoted the formation of spherical embryos;Cytokinins and its analogues played an important role in the genesis and maintenance of somatic embryo, their contents showed a tendency to first increase and then decrease, their contents were the lowest in EC stage and the highest in ICpEC stage, and tend to be stable in GE stage;gibberellins had a reverse regulation effect on the genesis of embryonic cells of S. emodi, in which GA3had a high content in NEC stage and the lowest content in EC stage, high concentrations of gibberellins inhibited the formation of somatic embryo, and showed a tendency to first increase and then decrease in 4 somatic embryogenesis stages of S. emodi;abscisic acid content showed a tendency to gradually decrease in 4 somatic embryogenesis stages of S. emodi, high concentrations of abscisic acid promoted the fast formation of somatic embryo of S. emodi, and the embryonic characteristic was maintained and the maturation of somatic embryo was delayed with the gradual decrease of abscisic acid concentration, which migh
