摘 要:目的 观察6 Gy60Coγ射线一次性全身照射对C57BL/6J小鼠血睾屏障的损伤,以及益气解毒方的防护效应。方法 80只C57BL/6J雄性小鼠根据体质量随机分为空白组、模型组、安多霖组(0.54 g/kg)和益气解毒方组(16.58 g/kg),每组20只。预防性给药10 d后,除空白组外,其余各组小鼠均给予6 Gy60Coγ射线一次性全身照射,并于照射后第4天持续给药至第11天。分别于照射后第2、12天处死小鼠,称量睾丸和附睾质量,HE染色观察睾丸及附睾病理变化、测量睾丸生精小管直径和生精上皮厚度,ELISA法检测血清抑制素B(INH B)水平,Western blotting检测小鼠睾丸中紧密连接蛋白-11及闭锁蛋白表达水平。结果 与空白组比较,模型组、安多霖组及益气解毒方组小鼠睾丸和附睾质量在照射后第2天差异无统计学意义,而照射后第12天均下降(P<0.01);模型组小鼠睾丸和附睾组织在照射后第2天即出现病理损伤,照射后第12天损伤加重,且在这2个时间点,生精小管直径和生精上皮厚度均较空白组下降(P<0.01)。益气解毒方组小鼠睾丸及附睾组织病理损伤均较模型组改善,且生精小管直径和生精上皮厚度较模型组均增加(P<0.01)。在照射后12 d,模型组小鼠血清INH B含量较空白组下降(P<0.05);益气解毒方组小鼠血清INH B含量较模型组上升(P<0.01)。照射后第2、12天,模型组小鼠睾丸中紧密连接蛋白-11、闭锁蛋白含量均较空白组下降(P<0.05,P<0.01),益气解毒方组小鼠睾丸中紧密连接蛋白-11、闭锁蛋白含量均较模型组上升(P<0.01)。结论 6 Gy60Coγ射线一次性全身照射导致雄性C57BL/6J小鼠血睾屏障严重损伤,益气解毒方能有效减轻该损伤,并促进生精功能的恢复,具有一定的防护效应。Objective To investigate blood-testis barrier injury in C57 BL/6 J mice induced by one-time total-body irradiation(TBI) with 6 Gy60Co γ ray, and the ability of Yiqi Jiedu decoction to protect against this injury.Methods Eighty male C57 BL/6 J mice were randomly assigned to the following groups according to body mass: blank group, model group, anduolin group(0.54 g/kg), and Yiqi Jiedu decoction group(16.58 g/kg), with 20 mice per group. After a 10-day pretreatment, the mice(except the blank group) were given one-time TBI with 6 Gy60Co γ ray, following which, treatment was administered from the 4thday to 11thday after radiation. The mice were sacrificed separately on the 2ndday and 12thday after radiation. The testis and epididymis were weighed, and their pathological changes were examined using HE staining. Additionally, the diameter of the seminiferous tubules and the thickness of the seminiferous epithelium were measured. In addition, the serum inhibin B(INH B) was detected using ELISA, and the expressions of claudin-11 and occludin in the testis were determined using Western blotting.Results Compared with the blank group, the weight of the testis and epididymis in the model group, anduolin group, and Yiqi Jiedu decoction group did not change significantly on the 2ndday after the TBI, yet all decreased on the 12thday after TBI(P < 0.01). The testis and epididymis in the model group showed pathological damages on the 2ndday that were aggravated on the 12thday, while the diameter of the seminiferous tubules and thickness of the seminiferous epithelium were decreased compared with those of the blank group at both time points(P<0.01). The pathological damages to the testis and epididymis in the Yiqi Jiedu decoction group were both improved when compared with those of the model group, and the diameter of the seminiferous tubules and thickness of the seminiferous epithelium increased compared with the model group at both time points(P<0.01).
