摘 要:植物源脲酶诱导碳酸钙沉淀(EICP)是近年来较为热门的新型土体固化技术,与微生物诱导碳酸钙沉淀(MICP)相比,具有简单易控制、环保更经济等优点。从黄豆、黑豆、绿豆和西瓜籽中提取脲酶,研究了脲酶浓度、尿素浓度、温度和pH值等4个因素对4种植物源脲酶活性的影响。在此基础上,研究了不同尿素与氯化钙浓度比与酶胶比对4种植物源脲酶碳酸钙沉淀量和沉淀率的影响规律。结果表明:相同条件下,黄豆脲酶的活性与诱导碳酸钙沉淀量和沉淀率最高;在各变量条件下,不同植物源脲酶的活性与诱导碳酸钙沉淀量和沉淀率均呈现出大致相同的变化规律。脲酶活性随脲酶浓度的增加而持续增长;随尿素浓度的增加先增长后减小,最优尿素浓度在1.3mol/L左右;在5℃~55℃范围内随温度的增加而大幅增长;随pH值的增加先增长后减小,黄豆、黑豆和绿豆脲酶的最优pH值为8,西瓜籽脲酶的最优pH值为7。碳酸钙沉淀量和沉淀率随浓度比的增加先增长后减小,最优浓度比为1∶1;当酶胶比大于1∶1后,碳酸钙沉淀量的增长速率明显降低;最大碳酸钙沉淀率对应的酶胶比为1∶1。Plant-derived urease induced calcium carbonate precipitation(EICP)is a popular new soil solidification technology in recent years.Compared with microbial induced calcium carbonate precipitation(MICP),it has the advantages of simple control,environmental protection and more economical benifit.Urease is extracted directly from soybean,black bean,mung bean and watermelon seeds.At first,the effects of urease concentration,urea concentration,temperature and pH on urease activity are studied.On this basis,the calcification test of four plant-derived ureases at different urea to calcium chloride concentration ratios and enzyme to gum ratios is carried out,and its influence law on the precipitation amount(rate)of calcium carbonate is discussed.The results show that the activity of soybean urease and the precipitation amount(rate)of inducing calcium carbonate are the highest under the same conditions,and the activity of different plant-derived urease and the precipitation amount(rate)of inducing calcium carbonate shows roughly the same change law under variable condition.Urease activity increases continuously with the increase of urease concentration,which increases first and then decreases with the increase of urea concentration,and the optimal urea concentration is about 1.3mol/L.Urease activity increases greatly with the increase of temperature in the range of 5~55℃,which increases first and then decreases with the increase of pH,and the optimum pH of soybean,black soybean and mung bean urease is 8,and the optimum pH of watermelon seed urease is 7.The precipitation amount(rate)of calcium carbonate increases first and then decreases with the increase of the concentration ratio,and the optimal concentration ratio is 1∶1.When the ratio of enzyme to gum is greater than 1∶1,the growth rate of precipitation amount of calcium carbonate decreases significantly,and the ratio of enzyme to gum corresponding to the maximum precipitation rate of calcium carbonate is 1∶1.
