摘 要:以1,8-桉叶油素型油樟2年生扦插苗1年生半木质化枝条为材料,对1,8-桉叶油素型油樟茎段组织培养进行研究。结果表明:5月份采集1,8-桉叶油素型油樟半木质化茎段,用质量浓度0.1%的HgCl 2消毒5 min,茎段的萌芽率最高,褐化率和污染率低;茎段萌芽最适培养基为MS+1.0 mg/L 6-BA+0.05 mg/L IBA,萌芽率为73.33%;最适增殖培养基为MS+1.0~2.0 mg/L 6-BA+0.05 mg/L IBA,增殖系数达到3.7;组培苗生根最适培养基为1/2 MS+0.5mg/L NAA,生根率达到70%。组培苗瓶内生根培养20~25 d,移入大棚内炼苗7~10 d,移栽到营养钵,移栽成活率可达80%以上。The stem tissue culture of C.longipaniculatum ct.1,8-eucalyptus was studied with annual semi-ligninized branches of C.longipaniculatum ct.1,8-eucalyptus to provide theoretical basis for rapid propagation of C.longipaniculatum nursery stock.The results show that the axillary bud germination rate of stem segments from semi-lignified stem segments of C.longipaniculatum ct.1,8-eucalyptus was the highest with 0.1%HgCl 2 disinfected for 5 min in the May,and the browning rate and pollution rate were low.The optimal medium for the germination of the stem segments of C.longipaniculatum ct.1,8-eucalyptus was MS+1.0 mg/L 6-BA+0.05 mg/L IBA,and the germination rate was 73.33%.The optimum medium for multiplication of plantlets of C.longipaniculatum ct.1,8-eucalyptus was MS+1.0~2.0 mg/L 6-BA+0.05 mg/L IBA,and the multiplication coefficient was 3.7.The suitable medium for rooting was 1/2 MS+0.5 mg/L NAA,and the rooting rate was 70.00%.After 20~25 days of rooting culture and 7 days of seedling training in plastic shed,after transplanted to a nutrition bowl,the survival rate of transplanting could reaches over 80%.
