摘 要:本研究以紫玉淮山带腋芽的茎段为外植体进行组培快繁技术探讨。结果表明:对紫玉淮山茎段的灭菌处理及诱导效果,以中上部茎段0.1%升汞处理4~8 min为宜,存活率高,污染率和褐化率低,灭菌效果好,同时诱导萌芽效果较好。初代培养基以MS+3 mg/L KT较好;继代腋芽培养增殖率最佳的培养基为MS+KT 3 mg/L+NAA 0.1~0.2 mg/L;生根培养基宜选用1/2 MS+NAA 0.5 mg/L;移栽基质为蛭石与营养土配比1:3。建立的组培快繁体系可为紫玉淮山工厂化生产种苗提供技术基础。In this study,stem segments with axillary buds of Dioscorea alata L.were used as explants for tissue culture and rapid propagation technology.The results showed that 0.1%HgCl2 treatment for 4-8 min in the middle and upper stems was suitable for sterilization treatment and induction effect with high survival rate,low pollution rate and browning rate,good sterili-zation,and good germination induction effect.MS+3mg/L KT made the best medium for primary culture.The medium with the best proliferation rate of subculture axillary buds was MS+KT 3 mg/L+NAA 0.1-0.2 mg/L.Rooting medium should be 1/2MS+NAA0.5mg/L.The transplanting substrate was vermiculite and nutrient soil ratio 1:3.The established tissue culture and rapid propagation system can provide technical basis for industrialized production of seedlings in D.alata L.
