柴芎头痛颗粒对偏头痛模型大鼠行为学及相关神经递质含量的影响

摘　　要：目的通过观察大鼠行为学与检测疼痛相关的神经递质含量,探讨柴芎头痛颗粒治疗偏头痛的药效学作用。方法试验动物随机分为正常对照组、模型对照组、阳性对照药正天丸3.20 g/kg组、柴芎头痛颗粒5.36、2.68、1.34 g/kg剂量组,共6组,每组12只大鼠。制备硝酸甘油型偏头痛大鼠模型。记录耳红开始及消失时间、挠头开始及消失时间;按Elisa试剂盒说明书测定各组大鼠血清中五羟色胺(5-HT)、5-羟基吲哚乙酸(5-HIAA)、β-内啡肽(β-EP)、多巴胺(DA)、一氧化氮(NO)水平。结果柴芎头痛颗粒高、中剂量组与模型组比较,可以明显延长抓耳开始时间、缩短耳红及挠头消失时间(P<0.01或P<0.05);高、中、低3个剂量组大鼠血清中5-HT、DA、β-EP含量显著升高、5-HIAA含量显著降低,与模型对照组比较差异明显(P<0.01或P<0.05);高、中剂量组可以显著降低大鼠血清中NO含量,与模型对照组比较差异明显(P<0.05)。结论柴芎头痛颗粒对实验性偏头痛模型大鼠具有显著镇痛作用,可以有效预防与治疗偏头痛。Objective To investigate the pharmacodynamic effect of Chaixiong Toutong Granules on migraine by observing the behavior of rats and detecting the content of pain-related neurotransmitters.Methods The experimental animals were randomly divided into a normal control group,a model control group,a positive control Zhengtian Pills group(3.20 g/kg),and Chaixiong Toutong Granules groups(5.36,2.68,1.34 g/kg,respectively),with 12 rats in each group.A nitroglycerin migraine rat model was established.The onset and disappearance time of ear redness and the onset and disappearance time of head scratching were recorded.The serum levels of serotonin(5-HT),5-hydroxyindoleacetic acid(5-HIAA),beta-endorphin(β-EP),dopamine(DA)and nitric oxide(NO)in each group were determined according to the instructions of Elisa kit.RReessuullttss Compared with the model group,the onset time of ear scratching and the disappearance time of ear redness and head scratching were significantly prolonged in the Chaixiong Toutong Granules high-and medium-dose groups(P<0.01 or P<0.05).The serum contents of 5-HT,DA andβ-EP in rats in the high-,medium-and low-dose groups were significantly increased,while the contents of 5-HIAA were significantly decreased,which were significantly different from those in the model control group(P<0.01 or P<0.05).The serum contents of NO in the high-and medium-dose groups were significantly decreased compared with that in the model control group(P<0.05).Conclusion Chaixiong Toutong Granules has a significant analgesic effect on experimental migraine model rats and can effectively prevent and treat migraine.
