摘  要:目的 基于“脾之液为涎”理论比较生枳壳和岭南特色炮制品制枳壳对健康大鼠唾液腺功能的影响,进一步探讨枳壳炮制缓和燥性的作用机制。方法 将30只雄性SD大鼠随机分为空白组、生枳壳低剂量组和高剂量组、制枳壳低剂量组和高剂量组,每日灌胃1次,持续给药4周,测定给药后各组大鼠饮水量、唾液流率、颌下腺指数,采用Elisa法检测大鼠颌下腺组织中血管活性肠肽(vasoactive peptide, VIP)、5-羟色胺(5-hydroxytryptamine, 5-HT)、P物质(substance P,SP)和水通道蛋白5(aquaporin5,AQP5)的含量,免疫组化法观察大鼠颌下腺AQP5的表达,HE染色观察颌下腺组织病理形态学变化。结果 与空白组比较,生枳壳组大鼠饮水量明显升高(P<0.01),唾液流率和颌下腺组织中VIP、5-HT和AQP5水平均明显降低(P<0.05,P<0.01);与生枳壳高剂量组比较,制枳壳组大鼠饮水量降低(P<0.01),唾液流率和颌下腺组织中VIP、5-HT含量与AQP5表达升高(P<0.05,P<0.01)。病理结果显示,生枳壳组大鼠颌下腺腺泡数量减少、大小不一、血管轻微扩张,而制枳壳组大鼠颌下腺组织形态有所改善。结论 生枳壳的燥性可表现为大鼠唾液腺生理功能的紊乱且作用效果呈剂量依赖性,制枳壳可缓和生枳壳的燥性,其作用机制可能与调控颌下腺神经肽VIP、5-HT的含量和AQP5的表达有关。Objective: To compare the effects of raw Fructus aurantii and processed Fructus aurantii on salivary gland function in healthy rats, and further explore the mechanism of processed Fructus aurantii on alleviating dryness. Methods: Thirty male SD rats were randomly divided into control group, low-dose group and high-dose group of raw Fructus aurantii, low-dose group and high-dose group of processed Fructus aurantii. They were administered with corresponding drugs by gavage for 4 weeks. After administration, the amount of drinking water, salivary flow rate and submandibular gland index of rats in each group were measured. The contents of VIP, 5-HT, SP and AQP5 in submandibular gland were detected by Elisa. The expression of AQP5 in submandibular gland was measured by immunohistochemistry, HE staining was used to observe the pathomorphological changes of submandibular gland. Results: Compared with the control group, the amount of drinking water in the raw Fructus aurantii group was increased significantly(P<0.01), the salivary flow rate and VIP, 5-HT and AQP5 in submandibular gland were decreased significantly(P<0.05, P<0.01). Compared with the high-dose group of raw Fructus aurantii, the amount of drinking water in the processed Fructus aurantii group was decreased(P<0.01), the salivary flow rate, the content of VIP, 5-HT and the expression of AQP5 in submandibular gland were increased(P<0.05, P<0.01). The pathological results showed that the number and size of submandibular gland acini in the raw Fructus aurantii group was decreased, and the blood vessels expanded slightly, while the histological morphology of submandibular gland in the processed Fructus aurantii group was improved. Conclusion: The dryness of raw Fructus aurantii can induce the disorder of physiological function of salivary gland in rats, and the effect is dose-dependent. The processed Fructus aurantii can alleviate the dryness of raw Fructus aurantii, and its mechanism may be related to the regulation of the content of VIP and 5-HT in submandibula
