摘  要:【目的】明确采自福州水稻基地中水稻的病毒种类,为生产上有效防控水稻病毒病提供科学依据。【方法】从福建省福州市仓山区水稻基地采集有矮缩、黄化、分蘖增多等症状的水稻样品共9株,采用RT-PCR方法对其病原进行鉴定。根据水稻草状矮化病RGSV P6和水稻锯齿叶矮缩病RRSV P10的基因序列分别设计一对特异性引物,提取感病水稻样品和健康水稻叶片的总RNA,并进行RT-PCR扩增和测序。【结果】在9株水稻样品中,均扩增到500 bp左右的片段,与RGSV预期片段大小一致;有1株同时扩增到750 bp左右的片段,与RRSV预期片段大小一致,而健康水稻中均未扩增到相关条带。【结论】水稻病毒田间的毒源种类比较多,也常常出现复合侵染和隐症现象,因此仅通过病害症状表现很难诊断病毒病,需借助分子生物学手段才能作出正确的诊断。RT-PCR鉴定结果表明采集的9株矮缩水稻样品均感染RGSV,有一株复合感染RRSV。水稻病毒病病原的鉴定将为水稻病害的防治提供参考依据。[Objective]This paper aims to identify the pathogen of rice dwarf diseases collected from fields in Fuzhou(Fujian Province),and to provide references for effective prevention and control of rice viral diseases.[Method]9 rice plants with stunting,chlorisis and excess tillering symptoms were gathered from the rice field in Fuzhou City.Reverse transcription PCR(RT-PCR)was used to detect two viruses from these rice plants.Two pairs of specific primers were designed based on the sequence of P6 gene of Rice grassy stunt virus(RGSV),and P10 gene of Rice ragged stunt virus(RRSV),respectively.Total RNA was extracted from sample rice leaves.And then specific bands were amplified and sequenced by RT-PCR.[Results]9 bands with about 500 bp was amplified in all rice samples,which was consistent with the expected fragment size of RGSV.A fragment with about 750 bp was amplified in one sample,which was consistent with the expected fragment size of RRSV,but no related band was detected from healthy rice.The fragments were confirmed by sequencing.[Conclusion]Viral diseases of rice may be co-infected by a variety of viruses,which would cause complex symptoms,syndromes or masking of symptoms.So,it was difficult to diagnose only by disease symptoms.Molecular biological technique was an effective way for the diagnosis of viral diseases.9 dwarf rice samples were all infected with RGSV,and one of them was co-infected with RRSV.The identification of the pathogen of rice virus diseases is useful to forecast and detect the viral diseases in fields and will provide references for the disease control.
