摘  要:以5-溴噻吩-2-甲醛和2-羟基-4-甲氧基苯乙酮为原料,经过羟醛缩合合成黄酮醇骨架,再分别与丙烯酰氯和4-硝基肉桂酸进行酯化,得到荧光探针HBNA和HBAC。采用核磁共振氢谱、碳谱和高分辨率质谱确证其结构。分别与半胱氨酸和同型半胱氨酸进行响应测试,发现HBNA对半胱氨酸和同型半胱氨酸响应具有差异性。HBAC在Cys浓度为0~400μmol/L范围内具有良好的线性关系,检测限为51 nmol/L。综上所述,本文制备的探针具有灵敏度高、对半胱氨酸和同型半胱氨酸区别性好、价格低廉等特点,为进一步研究区别两种氨基酸的探针提供借鉴。Using 5-bromothiophene-2-carbaldehyde and 2-hydroxy-4-methoxyacetophenone as raw materials,flavonol skeleton was synthesized by aldol reaction,and then esterified with acryyl chloride and 4-nitrocinnamic acid,respectively,to obtain fluorescent probe HBNA and HBAC.The structure was confirmed by~1H NMR,13C NMR and high resolution mass spectrometry.It was found that HBNA had different response to cysteine and homocysteine.HBAC has a good linearity in the range of 0~400μmol/L Cys concentration,and the detection limit is 51 nmol/L.In conclusion,the probes prepared in this paper have the characteristics of high sensitivity,good differentiation between cysteine and homocysteine,and low price,which can provide reference for further research on the differentiation of the two amino acids.
