摘  要:【目的】鉴定葡萄PLATZ(Plant AT-rich sequence and zinc-binding protein)家族转录因子,并探究其在种子发育过程中与IAA处理下的表达模式变化。【方法】基于葡萄基因组数据,通过多种生物信息学方法对葡萄PLATZ家族成员进行鉴定和分析,利用qRT-PCR分析其在葡萄种子不同发育时期及IAA处理下的表达模式。【结果】葡萄全基因组范围内共鉴定到13个PLATZ成员,分布于9条染色体上。系统进化分析结果表明,葡萄PLATZ蛋白成员分为4组(A、B、C、D),无其他物种聚类的E组成员。蛋白保守基序分析表明葡萄PLATZ蛋白具有较强保守性,内含子-外显子分布表明葡萄PLATZ基因在进化上具有多样性。同线性分析发现,VvPLATZ7~VvPLATZ8和VvPLATZ5~VvPLATZ11存在并联重复。顺式作用元件分析发现该基因家族的启动子包含生长发育、光响应、激素响应及胁迫响应相关作用元件。qRT-PCR结果分析表明,VvPLATZ5在无核葡萄种子发育过程中表达水平显著高于有核葡萄,VvPLATZ2和VvPLATZ6在有核葡萄种子发育过程中表达水平显著高于无核葡萄;8个VvPLATZ基因均响应IAA处理,且VvPLATZ5和VvPLATZ12基因在处理后6 h表达水平上调幅度显著。【结论】在葡萄基因组共鉴定到13个PLATZ基因,进化上同组成员保守基序数目及类别具有保守性,基因内含子-外显子结构具有多样性。PLATZ基因在有核、无核葡萄种子发育不同时期表达模式存在差异,且显著响应IAA处理,推测其在葡萄种子发育过程中发挥作用。【Objective】PLATZ is a plant-specific zinc finger transcription factor and has important functions in many processes of plant growth and development.It has also been reported to be involved in the development of seeds in other plants,but the regulatory mechanism in the seed of grape berry development is still unclear.By identifying the structure and expression of the grapevine PLATZ gene family,this study aimed to lay a foundation for further understanding the role of PLATZ family genes in various plant growth and developmental processes especially in seed development.【Methods】Based on the grape genome data,basic local alignment search tool(BLAST)and Hidden Markov Models(HMM)profiles were used to identify the members of the grape PLATZ family.Online websites including Pfam,NCBI,SMART,Ex PASy,PSORT,GSDS 2.0,MEME and bioinformatics analysis software including SPDE,MEGA-X and TBtools were used to predict the physicochemical properties,gene structure,protein subcellular localization,phylogenetic relationship and distribution of cis-acting elements of grape PLATZ gene family.qRT-PCR was used to analysis the expression level of the VvPLATZ genes during seed development and in response to exogenous IAA.【Results】A total of 13 PLATZ members were identified in the whole grape genome,which were distributed on 9 chromosomes.The amino acid sequence length of family members was between 115 and 306 aa.The maximum molecular mass was34700.11 u and the minimum was 12703.00 u.The isoelectric points were between 8.20 and 10.12,the instability factor was between 36.88 and 71.24,and the aliphatic index was between 62.37 and85.96.The subcellular localization prediction results showed that 9 of the 13 grape PLATZ family members were localized in the nucleus,2 in the cytoplasmic,1 in the chloroplast,and 1 in the peroxisome.The phylogenetic analysis showed that grape PLATZ protein members were divided into four groups(A,B,C,D),subgroups A,B,C,and D contain 2,3,6,and 2 grape PLATZ members,respectively,and subgroup did not contai
