摘 要:本文分析了实验各前处理阶段对饮用水中隐孢子虫和贾第鞭毛虫检测回收率的影响及损失程度。通过对定量抽滤、振荡淘洗、离心浓缩、免疫磁分离、染色镜检等不同前处理阶段的回收率进行测定,分析各阶段的回收率分布比例,并提出了新的统计指标——损失率。结果表明,隐孢子虫在各处理阶段的损失率依次为:30.7%(洗脱淘洗);23.0%(离心浓缩);17.0%(定量抽滤);4.3%(免疫磁分离);3.3%(染色镜检)。贾第鞭毛虫的损失率依次为:24.7%(洗脱淘洗);22.3%(离心浓缩);14.0%(定量抽滤);3.0%(免疫磁分离);2.3%(染色镜检)。隐孢子虫和贾第鞭毛虫在洗脱和离心两阶段的损失率合计分别高达53.7%和47.0%。由此可见,隐孢子虫和贾第鞭毛虫均是在洗脱和离心阶段的损失率最大,染色镜检阶段损失率最小。在饮用水卫生检测中,应着重优化前处理阶段中的洗脱和离心过程,从而显著提高实验质量。In this paper,the effects of different pretreatment stages on the recovery of Cryptosporidium and Giardia in drinking water and the degree of loss were analyzed.By measuring the recovery rate of different pretreatment stages such as quantitative suction filtration,oscillating elutriation,centrifugal concentration,immunomagnetic separation,staining microscopy,etc.,the distribution proportion of recovery rate in each stage is analyzed,and a new statistical index-loss rate is proposed.The results showed that the loss rate of Cryptosporidium in each treatment stage was 30.7%(elution and elution);23.0%(centrifugal concentration);17.0%(quantitative suction filtration);4.3%(immunomagnetic separation);3.3%(staining microscopy).The loss rate of Giardia was 24.7%(elution and elution);22.3%(centrifugal concentration);14.0%(quantitative suction filtration);3.0%(immunomagnetic separation);2.3%(staining microscopy).The total loss rates of Cryptosporidium and Giardia in the two stages of elution and centrifugation were as high as 53.7%and 47.0%,respectively.It can be seen that Cryptosporidium and Giardia have the highest loss rate in the elution and centrifugation stages,and the lowest loss rate in the staining microscopic examination stage.In drinking water sanitation testing,we should focus on optimizing the elution and centrifugation process in the pretreatment stage,so as to significantly improve the quality of the experiment.
