摘 要:构建美洲狼尾草(Pennisetum glaucum)的组织培养再生体系对于该物种的育种研究具有重要意义。以美洲狼尾草的种子、幼叶、幼穗和茎节作为外植体,研究这几种外植体的最佳消毒方式并挑选最合适的外植体进行组织培养,在组培体系中添加不同浓度的植物激素,研究其对愈伤组织诱导与分化的影响。结果表明:以种子作为外植体效果最好,5%NaClO消毒5 min后污染率为1.33%;诱导种子产生愈伤组织的最佳培养基为MS+3 mg·L^(-1)2,4-D+1 mg·L^(-1)6-KT+0.1 mg·L^(-1)6-BA,诱导率为93.3%;诱导胚性愈伤组织分化的最佳培养基为MS+3 mg·L^(-1)6-BA+1 mg·L^(-1)NAA,分化率为81.4%。The construction of tissue culture and regeneration system of Pennisetum glaucum is of great significance for the breeding of this species.In the present study,different explants of P.glaucum including seeds,tender leaves,young panicle,and stem node were subjected to disinfection and selected an appropriate explant for tissue culture.For this purpose,different plant growth regulators with different concentrations were applied to check the potential of callus induction,regeneration,and differentiation of each selected segment.The results showed that seed explant was the best compared to other explants regarding callus induction and differentiation,which were sterilized with 5%NaClO for 5 min(appropriate disinfection method)and found no contamination during callus formation.The highest rate of embryogenic callus induction(93.3%)was recorded in the MS medium when supplemented with 3 mg·L^(-1)2,4-D+1 mg·L^(-1)6-KT+0.1 mg·L^(-1)6-BA.Moreover,the highest differentiation rate of embryogenic callus(81.4%)was recorded in the MS medium at 3 mg·L^(-1)6-BA+1 mg·L^(-1) NAA.
