摘 要:目的利用网络药理学探讨茯苓桂枝对慢性心肌缺血的作用机制。方法经检测得到的成分在TCMSP平台筛选出活性成分。利用成分名在Pubchem平台找到成分对应SMILE结构。在Swiss Target Prediction平台获取成分作用的相关靶点。GeneCards数据库获取心肌缺血相关靶点。在Venny韦恩图平台作图。取药物作用靶点与心肌缺血相关靶点交集。Cytoscape软件作图“成分-药物-靶点-疾病”网络,使用String数据库绘制蛋白相互作用网络。在Biodnet数据库检索核心靶点对应的Gene ID号码。最后对核心靶点进行基因本体(GO)功能注释和(KEGG)通路富集。结果桂枝茯苓治疗心肌缺血具有多靶点多通路的特点,其机制可能与调控血管内皮细胞生长因子(VEGF)、低氧诱导因子(HIF-1)通路,非受体型酪氨酸激酶(SRC)、磷酸肌醇3-激酶调节亚基1(PIK3R1)、热休克蛋白90α编码基因(HSP90AA1)、丝/苏氨酸蛋白激酶(AKT)、丝裂原活化蛋白激酶(MAPK3)靶点有关,为进一步开展茯苓-桂枝治疗心肌缺血作用机制实验研究提供思路和依据。结论桂枝茯苓对心肌缺血具有调控作用。Objective To study the mechanism of action of Fulin-Guizhi on onchronic myocardial ischemia based on network pharmacology.Methods The active ingredients were screened out on the TCMSP platform from those components analyzed.Match the component names with their relative SMILE structrues on the Pubchem platform.The relevant targets of these components were then obtained on the Swiss Target Prediction Platform.The myocardial ischemia-related targets were obtained from the GeneCards databases,as well.Take the intersection of Therapeutic Drug Target and Myocardial ischemia related targets,by drawing the graph on Venny Platform.Draw the Ingredients-Targets-Drug-Disease network with the Cytoscape Software,and the String Database was used to draw the protein interaction network.Retrieve the gene ID number corresponding to the core targets in the biodnet database,and finally make the GO Functional Annotation and KEGG pathway enrichment analysis of the core targets.Results The treatment of myocardial ischemia by Fulin-Guizhi has the characteristic of multi-target and multi-channel.And its mechanism may have something to do with the regulation on the channel of VEGF,HIF-1 as well as the targets of SRC,IK3R1,HSP90AA1,AKT,MAPK3,which provides the idea and the support for the above research.Conclusion Fulin-Guizhi can regulate myocardial ischemia.
