摘 要:目的 研究壮骨关节丸中补骨脂素、异补骨脂素、补骨脂酚、川续断皂苷Ⅵ、蛇床子素、新补骨脂异黄酮、毛蕊花糖苷在大鼠血浆中的体内药动学。方法 8只大鼠灌胃给予该药物0.5%CMC-Na混悬液(5.4 g/kg),于给药前及给药后0.083、0.167、0.25、0.5、1、2、4、6、8、12、24 h采血,UHPLC-MS/MS法测定各成分血药浓度,计算主要药动学参数。结果 7种有效成分吸收较快,血药浓度在0.30~2.46 h达到峰值,其中补骨脂素、异补骨脂素、补骨脂酚暴露量更高。结论 该方法快速、准确、简便、重复性好,可为壮骨关节丸药效物质基础研究提供参考。AIM To study the in vivo pharmacokinetics of psoralen,isopsoralen,bakuchiol,aspersaponinⅥ,osthole,neobavaisoflavone and verbascoside of Zhuanggu Guanjie Pills in rat plasma.METHODS Eight rats were given intragastric administration of the 0.5% CMC-Na suspensions of this drug(5.4 g/kg),after which blood collection was made before administration and at 0.083,0.167,0.25,0.5,1,2,4,6,8,12,24 h after administration,UHPLC-MS/MS was adopted in the plasma concentration determination of various constituents,and main pharmacokinetic parameters were calculated.RESULTS Seven effective components demonstrated rapid absorption,whose plasma concentrations reached the peak at 0.30-2.46 h,among which psoralen,isopsoralen and bakuchiol displayed higher exposure.CONCLUSION This rapid,accurate,simple and reproducible method can provide a reference for the drug efficacy material base research on Zhuanggu Guanjie Pills.
