摘  要:目的:探讨石菖蒲中木脂素单体化合物Tatarinan O(TO)对转化生长因子β1(TGF-β1)诱导的非小细胞肺癌A459细胞上皮间质转化(EMT)及其迁移的影响。方法:通过TGF-β1诱导A549细胞EMT的发生。采用蛋白质印迹法、免疫荧光法检测E-钙黏蛋白(E-cadherin)、波形蛋白(Vimentin)、SMAD2蛋白和磷酸化SMAD2(p-SMAD2)蛋白的表达,以及SMAD2的核转位行为;采用划痕实验检测细胞迁移能力。结果:TO能够在A549细胞上抑制TGF-β1诱导的SMAD2的磷酸化,且呈剂量依赖性(P<0.05),并阻止其核转位。在25、50μmol/L的TO作用下,Vimentin表达明显降低,E-cadherin表达明显升高,与0μmol/L的TO相比,差异有统计学意义(P<0.05),并进一步抑制A549细胞迁移行为。结论:TO可通过SMAD2信号通路抑制TGF-β1诱导的非小细胞肺癌A549细胞的EMT,降低其迁移能力。OBJECTIVE:To investigate the effect of Tatarinan O(TO),a lignans monomeric compound in Acorus calamus,on transforming growth factor-β1(TGF-β1)-induced epithelial mesenchymal transition(EMT)and its migration of non-small cell lung cancer A459 cells.METHODS:The onset of EMT in A549 cells was induced by TGF-β1.The expression of E-cadherin,Vimentin,SMAD2 protein and phosphorylated SMAD2(p-SMAD2)protein,and the nuclear translocation behavior of SMAD2 were detected by using Western blotting and immunofluorescence assay;the cell migration ability was detected by scratch assay.RESULTS:TO was able to inhibit TGF-β1-induced phosphorylation of SMAD2 on A549 cells,which presented a dose-dependent(P<0.05)and prevented its nuclear translocation.The Vimentin expression was significantly reduced and the E-cadherin expression was significantly increased under the action of TO at 25 and 50μmol/L,with statistically significant differences(P<0.05)compared with TO at 0μmol/L,which further inhibited the migratory behavior of A549 cells.CONCLUSIONS:TO can inhibit TGF-β1-induced EMT in non-small cell lung cancer A549 cells through SMAD2 signaling pathway,and reduce its migratory ability.
