基于正交设计的西洋梨‘Conference’愈伤组织诱导研究

摘  要:以西洋梨‘Conference’为材料诱导愈伤组织,采用L9(34)正交设计法,探讨各因子对‘Conference’梨愈伤组织诱导效果的影响。结果表明,各因素对愈伤组织诱导率的影响表现为TDZ浓度>外植体类型>IBA浓度,TDZ、IBA浓度对试验结果的影响呈显著水平(P<0.05),不同水平间也差异显著。最优组合为A1B3C3,即以茎段为外植体,培养基和植物生长调节剂组合为MS+2.0 mg/L TDZ+0.5 mg/L IBA中暗培养35 d,诱导率可达100%。Pyrus communisL.‘Conference’was used as material to induce callus,and L9(3~4)orthogonal design was used to explore the effects of various factors on callus induction of‘Conference’pear.The results showed that the order of influence of each factor on callus induction:TDZ concentration>explants type>IBA concentration,and the influence of TDZ and IBA on the experimental results was significant(P<0.05),in addition,there were significant differences among different levels.A1B3C3proved to be the best combination,stem segments were used as explants,MS+2.0 mg/L TDZ+0.5 mg/L IBA medium and dark culture for 35 days were adopted,and the induction rate was 100%.
