摘 要:目的研究左归丸对卵巢早衰(POF)模型和初老大鼠下丘脑、垂体、卵巢组织中雌激素受体(ER)表达的影响,探讨左归丸促进卵巢功能恢复的作用机制。方法将60只青年SD雌性大鼠采用随机数字表法分为空白组、模型组、模型+补佳乐组、模型+左归丸低剂量组、模型+左归丸中剂量组、模型+左归丸高剂量组6组,每组10只。除空白组外,其余5组大鼠给予顺铂注射液每周6 mg/kg腹腔注射,共注射2次,建立POF大鼠模型。另取40只初老SD雌性大鼠,采用随机数字表法分为初老组、初老+左归丸低剂量组、初老+左归丸中剂量组、初老+左归丸高剂量组4组,每组10只。各组给予相应药物或生理盐水灌胃,连续灌胃21 d。灌胃结束后,采用酶联免疫吸附测定(ELISA)法检测各组大鼠血清卵泡刺激素(FSH)、促黄体生成素(LH)、雌二醇(E_(2))的水平,并通过免疫组化法和蛋白免疫印迹法检测各组大鼠下丘脑、垂体、卵巢组织中ER的表达水平。结果与空白组比较,模型组和初老组大鼠血清FSH水平明显升高,E_(2)水平明显降低,差异均有统计学意义(P<0.05或P<0.01);与模型组比较,模型+左归丸高剂量组大鼠血清FSH水平明显降低,E_(2)水平明显升高,差异均有统计学意义(P<0.05或P<0.01);与初老组比较,初老+左归丸高剂量组和初老+左归丸中剂量组大鼠血清LH水平明显降低,E_(2)水平明显升高,差异均有统计学意义(P<0.05或P<0.01)。与空白组比较,模型组和初老组大鼠下丘脑、垂体、卵巢组织中ER表达水平明显降低,差异均有统计学意义(P<0.05或P<0.01);给予高剂量左归丸干预后,POF模型和初老大鼠下丘脑、垂体、卵巢组织中ER表达水平升高(P<0.05或P<0.01)。结论左归丸可能通过升高下丘脑、垂体和卵巢组织中ER的表达水平,提高雌激素活性,从而调节下丘脑-垂体-卵巢轴的平衡,达到治疗和缓解POF的作用。Objective To study the effect of Zuogui Wan(左归丸)on estrogen receptor(ER)expression in hypothalamus,pituitary and ovarian tissue of premature ovarian failure(POF)model and primary senescence rats,and to explore the action mechanism of Zuogui Wan in promoting the recovery of ovarian function.Methods 60 young SD female rats were divided into blank group,model group,model plus progynova group,model plus low dosage Zuogui Wan group,model plus medium dosage Zuogui Wan group,model plus high dosage Zuogui Wan group using a random number table method,with 10 rats in each group.Except for the blank group,the other five groups were given intraperitoneal injection of cisplatin 6 mg/kg per week for 2 times to establish POF rat model.Another 40 primary senescence SD female rats were divided into four groups:primary senescence group,primary senescence plus Zuogui Wan low dosage group,primary senescence plus Zuogui Wan medium dosage group,and primary senescence plus Zuogui Wan high dosage group using a random number table method,with 10 rats in each group.Each group was given the corresponding medicine or normal saline by gastric irrigation for 21 consecutive days.After intragastric administration,the levels of serum follicle stimulating hormone(FSH),luteinizing hormone(LH)and estradiol(E_(2))were detected by enzyme-linked immunosorbent assay(ELISA)method.The expression levels of ER in hypothalamus,pituitary and ovarian tissues of rats in each group were detected by immunohistochemistry and protein immunoblotting.Results Compared with the blank group,the serum FSH levels in the model group and the primary senescence group rats were increased significantly,and the E_(2)levels were decreased significantly,the differences were both statistically significant(P<0.05 or P<0.01).Compared with the model group,the serum FSH level in the model plus high dosage Zuogui Wan group was decreased significantly,and the E_(2)level was increased significantly,the differences were statistically significant(P<0.05 or P<0.01).Compared with the
