摘 要:大熊猫Ailuropoda melanoleuca的肠道微生物十分复杂,其菌群结构直接关系到大熊猫的健康。本文采用5种商业化益生菌培养基(GYP、LBS、M17、SL和TJA)培养大熊猫粪便样品菌悬液24 h,用气相色谱法测定7种短链脂肪酸(SCFA)的含量。结果表明,SL和LBS中的SCFA含量最高,浓度分别达到16.410μg·mL-1和15.650μg·mL-1,其中乙酸占总含量的97%,而其他6种甚微,说明这2种培养基对产乙酸肠道菌有较强的富集作用。M17中的SCFA总含量虽然不高(2.410μg·mL-1),但可以检测到7种SCFA,正丁酸最高(1.230μg·mL-1),约占总含量的51%,丙酸和正己酸含量也占10%以上,说明M17培养基可以富集大熊猫肠道中较多种类的产酸菌。研究结果为大熊猫肠道益生菌的富集培养和分离筛选提供了基础资料。The gut microbes of giant pandas(Ailuropoda melanoleuca)are very complex,and the structure of their flora is directly related to the health of giant pandas.In this paper,bacterial suspensions of giant panda feces samples were cultured in 5 commercial probiotic culture media,including GYP,LBS,M17,SL and TJA,for 24 h.Subsequently,the contents of 7 short-chain fatty acids(SCFAs)were determined by gas chromatography.The results showed that the contents of SCFAs in SL and LBS culture media were the highest,and the concentrations reached 16.410μg·mL-1 and 15.650μg·mL-1,respectively.The content of acetic acid accounted for 97%of the total,and the other 6 SCFAs had very little content,indicating that these 2 media contributed to selecting acetogenic intestinal bacteria.Although the total amount of SCFAs in M17 culture medium was not high(2.410μg·mL-1),all the 7 SCFAs could be detected,and the concentration of n-butyric acid was the highest,reaching 1.230μg·mL-1,accounting for about 51%of the total content.The content of acid and n-hexanoic acid also accounted for more than 10%,indicating that M17 culture medium can enrich more types of acid-producing bacteria in the intestine of giant pandas.The results of this study lay a foundation for the enrichment,culture,isolation and screening of intestinal probiotics in giant pandas.
