摘  要:目的探讨长链非编码RNA(lncRNA)与七氟烷麻醉副作用的关联。方法采用小鼠模型分析七氟烷对海马锥体的影响,采用生物信息学方法分析GEO数据库中七氟烷处理的小鼠相关的lncRNA表达谱数据,筛选可能与七氟烷麻醉过程相关的lncRNA,并使用qPCR对lncRNA进行验证。结果七氟烷处理可导致小鼠海马锥体神经元结构异常,增加细胞凋亡。基因芯片分析显示,在小鼠(GSE155770)中,七氟烷处理后27个lncRNA出现差异表达。qPCR验证发现,经七氟烷处理后,小鼠海马组织lncRNA Gm11525和Gm19439的表达水平提高。结论不适量的七氟烷处理可引起海马锥体神经元结构改变和细胞凋亡,lncRNA可能参与了该病理性作用。Objective To explore the relationship between long noncoding RNA(lncRNA)and the anesthetic side effect induced by sevoflurane.Method C57BL/6 mouse were used to evaluate the influence of sevoflurane on hippocampal pyramidal neuron.lncRNA expression in mice was analyzed from GEO database and lncRNA related to sevoflurane were selected.Screened lncRNA were validated by using qPCR in mice.Result A single 6 h sevoflurane exposure to six-week-old female mice resulted in significant morphological changes and apoptosis in the hippocampus.From GEO database,27 significant difference in expression of lncRNA induced by sevoflurane were identified in GSE155770.We tested 5 of those 27 lncRNA by qPCR and found that lncRNA Gm11525 and Gm19439 were significantly up-expressed in sevoflurane exposed group than control group(exposed to air).Conclusion Sevoflurane exposure results in significant morphological changes and apoptosis in the hippocampus in mice and the lncRNA may involve in the mechanism of sevoflurane.
