摘 要:目的铁蛋白与许多肿瘤有关,但其机制仍不清楚。本研究旨在研究铁蛋白重链(ferritin heavy chain,FTH)和铁蛋白轻链(ferritin light chain,FTL)在前列腺癌中的作用以及调节前列腺癌PC-3细胞增殖和迁移的蛋白。方法进行集落形成和Transwell测定以评估稳定FTH沉默细胞系(stable FTH-silenced cell lines,LV-shFTH)、稳定FTL沉默细胞系(stable FTL-silenced cell lines,LV-shFTL)和阴性对照(negative controls,NC)的增殖和迁移功能。采用iTRAQ(isobaric tags for relative and absolute quantitation)和生物信息学分析用于检测蛋白质组学。结果与NC组相比,FTH沉默的细胞增殖和迁移能力降低,而FTL沉默的细胞则相反。在LV-shFTH和NC组之间共鉴定出442种上调蛋白和420种下调蛋白。LV-shFTL和NC组之间共鉴定出171种上调蛋白质和423种下调蛋白质。其中,有21个蛋白在LV-shFTH组和LV-shFTL组与NC组变化方向相反。结论FTH和FTL在PC-3细胞增殖和迁移中可能发挥不同的作用。此外,FTH和FTL通过21种蛋白质的差异表达影响PC-3细胞的增殖和迁移。Objective Ferritin is known to be associated with numerous tumors,but its mechanism remains to be elucidated.The present study aims to identify the role of ferritin heavy chain(FTH)and ferritin light chain(FTL)in prostate cancer and the mediators that regulate PC-3 cell proliferation and migration.Methods Colony formation and Trans-well assays were performed to evaluate the function of stable FTH-silenced cell lines(LV-shFTH),stable FTL-silenced cell lines(LV-shFTL)and negative controls(NC).Isobaric relative and absolute quantitative proteomics and bioinformatics analysis methods were used to detect their proteomics.Results Compared with the NC group,FTH-silenced cells showed reduced cell proliferation and migration abilities,while FTL-silenced cells showed the opposite trends.A total of 442 upregulated proteins and 420 downregulated proteins were identified between the LV-shFTH and NC groups.Additionally,171 upregulated proteins and 423 downregulated proteins were identified by analyzing the changes in protein expression profiles between LV-shFTL and NC groups.Among them,21 proteins were changed in opposite directions in the LV-shFTH group and the LV-shFTL group with the NC group.Conclusion FTH and FTL could exert different roles in PC-3 cell proliferation and migration.In addition,FTH and FTL affect PC-3 cell proliferation and migration via differential expressions of the identified 21 proteins.
