摘 要:目的研究含氯消毒剂对病毒RNA的消除效率,对其影响因素进行分析。方法以纯化流感病毒RNA作为研究对象,对前期研究有效的含氯消毒剂,基于化学成分、作用浓度及时间、载体特性等因素进行分析,以荧光定量聚合酶链反应(PCR)反映消除效率。结果经中和剂鉴定试验选用含0.5%硫代硫酸钠的磷酸盐缓冲液(PBS)中和残留消毒剂后,实验结果表明以有效氯含量为500 mg/L含氯消毒剂可实现对RNA的完全清除,且不同化学成分、不同类型载体对消毒剂清除效率的影响较小,进一步降低作用浓度至10 mg/L,经浸泡作用5 min也能实现对RNA污染的有效清除。模拟现场测试表明,玻璃、塑料和桌面上的RNA污染均可经1000 mg/L含氯消毒剂以擦拭或喷洒消毒的方式去除,而仅以物理擦拭或冲刷的方式未能清除RNA污染。结论与物理擦拭方式相比,含氯消毒剂充分接触作用后对纯化的病毒RNA具有很好的消除效果。Objective To study the elimination efficiency of chlorine-containing disinfectants on viral RNA and analyze its influencing factors.Methods Purified influenza virus RNA was taken as the research object in this study.Based on different factors such as chemical composition,concentration,duration of action and carrier characteristics,fluorescence quantitative PCR was used to evaluate the effect of previous research of chlorine-containing disinfectants on the viral RNA elimination.Results The neutralizer identification test showed that PBS containing 0.5%sodium thiosulfate was selected to neutralize the residual disinfectant.The experiment showed that the disinfectant with available chlorine content of 500 mg/L could remove RNA completely,and different chemical composition and different types of carriers had little effect on the removal efficiency.Further study showed removal of RNA could also be achieved with chlorine-concentration of 10 mg/L after 5 minutes.Field tests of simulated contamination showed that RNA contamination on glass,plastic and tabletops could be removed by wiping or spraying with 1000 mg/L chlorine-containing disinfectant,while physical wiping and scouring failed to remove RNA contamination.Conclusions Compared with the physical method,chlorine-containing disinfectant has a good effect of elimination on viral RNA.
