摘 要:目的探讨灰树花多糖对乳腺癌细胞放疗敏感性的影响及机制。方法50、100、200、400 mg·L^(-1)的灰树花多糖处理MDA-MB-231细胞24 h和48 h,采用MTT法检测细胞增殖。克隆形成实验检测400 mg·L^(-1)的灰树花多糖对MDA-MB-231细胞放射增敏效果。将MDA-MB-231细胞分为对照组、灰树花多糖组、照射组和联合组。流式细胞术检测细胞周期和细胞凋亡率。DCFH-DA活性氧检测试剂盒检测ROS水平。Western blotting检测Notch1、Hes1、cyclinE1和cleaved caspase 3表达。结果灰树花多糖可呈时间浓度依赖性抑制MDA-MB-231细胞增殖(P<0.05)。由于400 mg·L^(-1)的灰树花多糖处理MDA-MB-231细胞24 h可抑制近一半的细胞增殖,因此选择作为研究浓度。灰树花多糖联合照射细胞存活分数明显低于单纯照射组(P<0.05)。灰树花多糖和照射均可明显提高G0/G1细胞百分比,降低S期细胞百分比,促进细胞凋亡和ROS产生,下调Notch1、Hes1和cyclinE1表达,上调cleaved caspase 3表达(P<0.05),灰树花多糖与照射联合使用对MDA-MB-231细胞周期、凋亡、ROS水平及Notch1、Hes1、cyclinE1和cleaved caspase 3表达的影响更明显,且与灰树花多糖组或照射组比较差异有统计学意义(P<0.05)。结论灰树花多糖可抑制乳腺癌细胞增殖,阻滞细胞周期,促进细胞凋亡,且可增加放疗敏感性,机制可能与诱导ROS产生及抑制Notch1信号通路有关。Objective To investigate the effect of Grifola frondosa polysaccharide on radiosensitivity of breast cancer cells.Methods MDA-MB-231 cells were treated with 50,100,200 and 400 mg·L^(-1)Grifola frondosa polysaccharides for 24 h and 48 h.The proliferation of MDA-MB-231 cells was detected by MTT assay.The radiosensitization effect of Grifola frondosa polysaccharide(400 mg·L^(-1))on MDA-MB-231 cells was detected by clone formation assay.MDA-MB-231 cell was divided into control group,Grifola frondosa polysaccharide group,irradiation group and combination group.Cell cycle and apoptosis rate were detected by flow cytometry.The level of ROS was detected by DCFH-DA reactive oxygen species detection kit.Western blotting was used to detect the expressions of Notch1,Hes1,cyclinE1 and cleaved caspase 3.Results Grifola frondosa polysaccharide inhibited the proliferation of MDA-MB-231 cells in a time and concentration-dependent manner(P<0.05).Since 400 mg·L^(-1)Grifola frondosa polysaccharide treatment MDA-MB-231 cells for 24 hours can inhibit nearly half of the cell proliferation,it was chosen as the study concentration.The survival fraction of Grifola frondosa polysaccharide combined with irradiation group was significantly lower than that of irradiation group alone(P<0.05).Grifola frondosa polysaccharide and irradiation could significantly increase the percentage of G0/G1 cells,reduce the percentage of S phase cells,promote apoptosis and ROS production,down-regulate the expressions of Notch1,Hes1 and cyclinE1,and up-regulate the expression of cleaved caspase 3(P<0.05).The combined use of Grifola frondosa polysaccharide and irradiation had more significant effects on MDA-MB-231 cell cycle,apoptosis,ROS level and the expressions of Notch1,Hes1,cyclinE1 and cleaved caspase 3,and the differences were statistically significant compared with Grifola frondosa polysaccharide group or irradiation group(P<0.05).Conclusion Grifola frondosa polysaccharide can inhibit the proliferation of breast cancer cells,block cell cycle,promote c
