摘 要:目的通过建立坏死性小肠结肠炎的小鼠模型,检测新生子鼠发生NEC早期时血清中细胞焦亡的关键蛋白分子,从而初步判断细胞焦亡是否参与早期新生儿坏死性小肠结肠炎的发病。方法将新出生的子鼠随机分为实验组和对照组,实验组的子鼠诱导为NEC的动物模型,抽取2组子鼠血液,用Elisa试剂盒检测所有子鼠血清中IL-18、Gasdermin D、Caspase-1的浓度,运用秩和检验统计学方法,比较2组血清中目标蛋白的浓度,检验水准α=0.05。结果2组子鼠血清中Gasdermin D、Caspase-1的浓度差异有统计学意义,P值分别为0.017、<0.001,可认为实验组子鼠血清中Gasdermin D、Caspase-1的浓度高于对照组,2组血清中IL-18的浓度无明显差异,P=0.755。结论由Gasdermin D介导的细胞焦亡可能参与早期NEC的发病。Objective To establish a mouse model of necrotizing enterocolitis and detect the key protein molecules in serum of neonatal rats with early NEC,so as to preliminatively determine whether necrotizing enterocolitis is involved in early neonatal necrotizing enterocolitis.Methods The NEC mouse model was established and randomly divided into the experimental group and control group with 12 newborn rats in each group.The neonatal rats in the experimental group were induced into NEC animal models.Blood samples were extracted and the concentrations of IL-18,Gasdermin D and Caspase-1 in serum of all rats were detected by Elisa kit.The concentration of target protein in serum of the two groups was compared,α = 0.05.Results The concentrations of Gasdermin D and Caspase-1 in serum of rats in the two groups were significantly different,with P values of 0.017and < 0.001,respectively.It could be considered that the concentrations of Gasdermin D and caspase-1 in serum of rats in the experimental group were higher than those in the control group.There was no significant difference in serum IL-18 concentration between the two groups(P = 0.755).Conclusion Gasdermin D mediated pyroptosis may be involved in the pathogenesis of early NEC.
