摘 要:为探究外膜蛋白基因ompA对鼠伤寒沙门菌的生物学特性及对毒力的影响,本研究利用λ-Red同源重组技术构建ompA基因缺失株,通过比较检测生长曲线、运动性、生化特性、生物被膜形成能力、耐药性、细胞粘附性以及半数致死量(LD50)来检测野生株与基因缺失株之间的区别。结果显示,野生株与ompA缺失株的生长曲线、运动性、生化特性无明显区别;ompA缺失株的生物被膜形成能力极显著下降,对多种抗生素的敏感性增加,对细胞的粘附性和侵袭力均下降约79%;LD50的测定结果显示,STM LT2的LD50显著低于ompA缺失株,说明ompA的缺失显著降低了沙门菌对小鼠的致病力。综上所述,ompA的缺失能影响鼠伤寒沙门菌的部分生物学特性。以上结果为进一步研究外膜蛋白基因ompA基因的生物学功能及沙门菌致病性奠定了理论基础。In order to explore the biological characteristics and virulence of outer membrane protein gene ompA on Salmonella typhimurium, λ-Red homologous recombination technology was used in this study to construct ompA gene deletion strains. The following parameters were used to compare ompA deletion strain and parent strain, including the growth curve, motility, biochemical property,biofi lm-forming ability, drug resistance, cell adhesion, and median lethal dose(LD50). The results showed that the growth curves, motility and biochemical characteristics were not signifi cantly different between the wild strain and ompA-defi cient strain. Compared with STM LT2, the biofilm formation ability of the ompA-deficient strain significantly decreased while the susceptibility to various antibiotics increased. The adhesion and invasiveness to cells also decreased by about 79%. In addition, the LD50 of STM LT2 was signifi cantly lower, indicating the deletion of ompA signifi cantly reduced the pathogenicity to mice. Taken together, deletion of ompA affected some biological characteristics of S. typhimurium. These results laid a solid foundation for further research on the biological function of the outer membrane protein gene ompA and the pathogenicity of S. typhimurium.
