摘 要:调查云南锡金小鼠体内是否携带鼠疫噬菌体,并探讨其流行病学意义。2016-2017年,在云南鼠疫疫源地梁河县、宜良县及文山市3个调查点进行锡金小鼠的捕获,取其肠道标本,以鼠疫疫苗株EV76为宿主菌,采用双层平板法进行鼠疫噬菌体的分离,所分离噬菌体进行噬菌斑及电镜观察;同时进行鼠疫菌特异基因caf1的检测。结果共捕获86只锡金小鼠,从中分离到3株鼠疫噬菌体,分离率为3.49%;其噬菌斑在双层平板上表现为特大(直径>3.0mm)、中(~1.0mm)及小(<0.5mm)3种噬菌斑;对3株噬菌体进行电镜观察,皆为肌尾病毒科噬菌体;caf1基因检测皆为阴性。在家鼠鼠疫疫源地锡金小鼠中存在鼠疫噬菌体,锡金小鼠在鼠疫菌保存及远距离传播中的作用及意义值得进一步探讨。To explore whether Yersinia pestis phages were carried in Mus pahari in Yunnan province and its relationship with the plague epidemic, the rodents were captured from 2016 to 2017 in 3 places of Lianghe county, Yiliang county and Wenshan city and their intestinal specimens were taken for isolation of phages. The Y. pestis vaccine strain EV76 was used as the feeding bacteria and the twolayer plate method was used for phage solation. The isolated phages were examined under the electric microscope. At the same time, the specifi c gene caf1 of Y. pestis was tested in the isolated phages. A total of 86 Mus pahari were captured and 3(3.49%) Y. pestis phages were isolated. These phages had different sizes(large>3.0 mm, medium 1.0 mm and small<0.5 mm in diameter) on the double-layer plate. Two phages belonged to be myoviridae as observed under electron microscope. Three phages were all caf1 negative. This study demonstrated the presence of Y. pestis phages in Mus pahari in these places. The role and epidemiological signifi cance of Mus pahari in the preservation of plague is needed for further investigation.
