摘 要:为了解我国最新流行的鸽新城疫病毒(Newcastle disease virus,NDV)分子生物学特征,对2021-2022年国家新城疫参考实验室从不同地区鸽群中分离到的8株NDV代表株进行了全基因组测序和生物信息学分析。结果显示:8株鸽NDV基因组长度均为15 192 nt,基因排列方式为3’-NP-P-M-F-HN-L-5’,其F蛋白裂解位点氨基酸序列均为112RRQKR/F117,呈典型的NDV强毒分子特征;相较于参考序列,代表毒株F蛋白七肽重复区域、融合肽和跨膜区域,以及HN蛋白中和抗原表位、跨膜区域等功能区域均有多处氨基酸变异;代表毒株全基因组核苷酸同源性为91.9%~99.0%,均属于Class Ⅱ基因Ⅵ型病毒,但可分为2个小分支,其中4株属于基因Ⅵ.2.1.1.2.2亚型,另4株属于基因Ⅵ.2.1.1.2.1亚型。结果表明,我国近期流行的鸽NDV具有遗传多样性特征。本研究进一步掌握了我国鸽NDV的遗传进化特点,丰富了鸽新城疫的流行病学信息,为该病的科学防控提供了参考。In order to investigate the molecular biological characteristics of pigeon Newcastle disease virus(NDV)prevalent in China in recent years,whole genome sequencing and bioinformatics analysis were conducted on 8 strains of NDVs isolated from pigeons in different regions by National Reference Laboratory of Newcastle Disease from 2021 to 2022. The results showed that the genome length of all the strains were 15 192 nt,with the gene order of 3’-NP-P-M-F-HN-L-5’,and all their cleavage sites of F proteins were 112RRQKR/F117,with typical characteristics of virulent strain;compared to the reference sequence,multiple amino acid mutations were observed in heptad repeat region,fusion peptide and transmembrane region of F protein of the representative strains,and the neutralizing epitope and transmembrane domain of HN protein;the nucleotide homology of the whole genomes of the 8 strains and the representative strain was from 91.9% to 99.0%,all of which belonged to type Ⅵ of Class Ⅱ branch,but could be divided into two sub-branches,four of which belonged to Ⅵ.2.1.1.2.2 subtype,and the other four belonged to Ⅵ.2.1.1.2.1. In conclusion,genetic diversity was available in pigeon NDV prevalent in China. The genetic evolution characteristics of pigeon NDV in China were identified by the study,which enriched relevant epidemiological information,and provided a reference to scientifically prevent and control the disease.
