摘 要:目的:探索不同生态环境对东方泽泻生理活动及其药效成分原萜烷型三萜生物合成调控的分子基础。方法:通过Illumina HiSeq测序平台获取种植于福建和四川的东方泽泻块茎转录组数据并进行功能注释,基于|log_(2)FC|>0.58(FC为差异倍数)、P<0.05的筛选条件获得差异表达基因。结果:转录组测序共获得50652条unigenes,有22689条unigenes注释到京都基因与基因组百科全书(KEGG)、NR、SwissProt、COG、基因本体(GO)和Pfam数据库。从福建和四川2种环境生长的东方泽泻之间筛选到3878条差异表达基因,其中2082条在福建东方泽泻中上调表达,1796条在四川东方泽泻中上调表达,差异基因主要参与植物防御反应、光合作用、淀粉和蔗糖代谢、萜类生物合成等生理活动,包括5个原萜烷型三萜生物合成通路上的关键酶基因。此外,东方泽泻移栽四川后,与三萜生物合成相关的转录因子和细胞色素P450(CYP450)基因表达也发生了显著变化。结论:利用高通量测序技术和生物信息分析获得了不同生态环境下东方泽泻转录组信息特征,为阐明生态环境对东方泽泻原萜烷型三萜生物合成的调控机制提供参考。Objective:To explore the molecular mechanism of different ecological environments on the regulation of physiological activities and biosynthesis of protostane triterpenes of Alisma orientale.Methods:The transcriptome data of A.orientale tubers from Fujian and Sichuan were obtained by Illumina HiSeq sequencing platform,followed by functional annotation,and differentially expressed genes(DEGs)were obtained based on the screening conditions of|log_(2)FC|>0.58 and P<0.05.Results:A total of 50652 unigenes were obtained by transcriptome sequencing,and 22689 unigenes were annotated to Kyoto Encyclopedia of Genes and Genomes(KEGG),NR,SwissProt,COG,Gene Ontology(GO),and Pfam.A total of 3878 DEGs were screened out from A.orientale grown in Fujian and Sichuan,including 2082 with up-regulated expression in Fujian and 1796 with up-regulated expression in Sichuan.DEGs were mainly involved in physiological activities such as plant defense response,photosynthesis,starch and sucrose metabolism,and terpene biosynthesis,including five key enzyme genes involved in the biosynthesis of protostane triterpenes.In addition,the expression of transcription factors and CYP450 genes related to triterpene biosynthesis also changed significantly after A.orientale transplantation to Sichuan.Conclusion:The high-throughput sequencing technology and bioinformatics analysis were used to obtain the transcriptome information characteristics of A.orientale under different ecological environments,which laid a foundation for clarifying the regulation mechanism of ecological environment on the biosynthesis of protostane triterpenes of A.orientale.
