摘  要:目的 探讨改良快速骨组织脱钙法在透射电镜样品制备中的应用效果。方法 使用JYBL-Ⅱ组织脱钙液,加入电镜专用戊二醛固定液,并对渗透压等影响因素进行了有效控制,与传统脱钙法处理电镜样品后进行比较。结果 改良快速骨组织脱钙法使脱钙时间由传统的2~4周缩短至24 h,电镜观察骨组织超微结构完整性更好,结构更清晰。结论 改良快速骨组织脱钙法在透射电镜样品制备中具有较高的应用价值,值得推广。Objective To investigate the application of improved rapid bone decalcification method in the sample preparation of transmission electron microscopy. Methods The improved method used JYBL-Ⅱ tissue decalcification solution and added glutaraldehyde fixative solution for electron microscope. The influence factors such as osmotic pressure were effectively controlled. The results were compared with those of traditional decalcification method. Results The improved rapid bone decalcification method shortened the decalcification time from the traditional 2-4 weeks to 24 hours, and the ultrastructure of bone tissue is more complete and clearer under electron microscopy. Conclusion The improved method of rapid bone decalcification has important application value in TEM sample preparation.
