摘 要:目的基于气道重塑关键信号通路ERK/MAPK研究针刺大椎、肺俞、足三里穴位对慢性哮喘小鼠气道重塑的影响机制。方法腹腔注射卵清蛋白(OVA)+Al(OH)3建立慢性哮喘小鼠模型,针刺大椎、肺俞、足三里穴位干预。将动物随机分成5组:PBS组(对照组)、OVA组(哮喘组)、OVA+Acu组(针刺治疗组)、OVA+PD98059组(抑制剂组)、OVA+Sham组(假针刺组)。使用ELISA法检测小鼠血清OVA特异性IgE水平,HE染色检测气道炎症程度,Masson染色和PAS染色检测气道重塑程度,Western blot检测小鼠肺组织上皮间质转化(EMT)相关蛋白和ERK/MAPK通路关键蛋白表达情况。结果ELISA结果显示,与OVA组相比,OVA+Acu组和OVA+PD98059组血清特异性IgE表达水平降低(均P<0.05)。HE染色结果显示,与OVA组相比,OVA+Acu组和OVA+PD98059组支气管周围炎性细胞浸润减少。Masson染色结果显示,与OVA组相比,OVA+Acu组和OVA+PD98059组支气管周围胶原沉积减少。PAS染色结果显示,与OVA组相比,OVA+Acu组和OVA+PD98059组杯状细胞增生减少。Western blot结果显示,与OVA组相比,OVA+Acu组和OVA+PD98059组E-cadherin蛋白表达增多(均P<0.05),OVA+Acu组(均P<0.05)和OVA+PD98059组(均P<0.01)Vimentin及α-SMA蛋白表达减少。此外OVA+Acu组(P<0.01)和OVA+PD98059组(P<0.05)相比于OVA组p-ERK蛋白表达减少。结论针刺大椎、肺俞、足三里可以改善慢性哮喘小鼠气道重塑,其作用机制可能与抑制ERK/MAPK信号通路的激活相关。Objective To explore the effects of acupuncture at Dazhui,Feishu and Zusanli on airway remodeling in chronic asthmatic mice based on ERK/MAPK.Methods A chronic asthma model was established by intraperitoneal injection of OVA+Al(OH)3,and acupuncture at Dazhui,Feishu and Zusanli was performed for intervention.The animals were randomly divided into 5 groups:PBS group(normal group),OVA group(asthma group),OVA+Acu group(acupuncture treatment group),OVA+PD98059 group(inhibitor group),and OVA+Sham group(sham acupuncture group).The level of OVA-specific IgE in serum was detected by ELISA,the degree of airway inflammation was detected by HE staining,the degree of airway remodeling was detected by Masson staining and PAS staining,and the epithelial-mesenchymal transition(EMT)-related proteins and key proteins in ERK/MAPK pathway in lung tissue were detected by Western blotting.Results Compared with the OVA group,the OVA-specific IgE expression levels in the OVA+Acu group and OVA+PD98059 group were decreased(P<0.05;P<0.05);compared with the OVA group,the infiltration of inflammatory cells in the OVA+Acu group and OVA+PD98059 group was reduced;compared with the OVA group,the collagen deposition in the OVA+Acu group and OVA+PD98059 group was reduced;compared with the OVA group,epithelial goblet cell proliferation in OVA+Acu group and OVA+PD98059 group was decreased;compared with the OVA group,the expression of E-cadherin protein in OVA+Acu group and OVA+PD98059 group increased(P<0.05;P<0.05),Vimentin(P<0.05;P<0.01)andα-SMA Protein expression decreased(P<0.05;P<0.01).The expression of p-ERK protein in OVA+Acu group and OVA+PD98059 group was decreased compared with OVA group(P<0.01;P<0.05).Conclusion Acupuncture at Dazhui,Feishu and Zusanli can improve airway remodeling in chronic asthmatic mice.The mechanism may be related to inhibiting the activation of ERK/MAPK signaling pathway.
