摘 要:目的探讨人类卵裂期胚胎和囊胚的不同形态学分型与自身线粒体分布和线粒体膜电位(mitochondrial transmembrane potential,ΔΨm)的关系。方法选择在生殖中心因女性双侧输卵管梗阻行体外受精-胚胎移植(in vitro fertilization-embryo transfer,IVF-ET)助孕顺利娩出健康胎儿后,自愿丢弃冷冻的卵裂期胚胎和囊胚用于研究。将冷冻的胚胎解冻复苏后,依据胚胎发育的时期,将其分为D3冷冻胚胎组和D5冷冻胚胎组,再根据冷冻胚胎的形态学评分标准,将D3冷冻胚胎组及D5冷冻胚胎组分别分为优质胚胎组和较差胚胎组。采用Mito-Tracker Green荧光探针检测胚胎线粒体在胚胎内的分布情况,采用荧光探针JC-10检测胚胎线粒体ΔΨm,比较相同阶段不同质量的胚胎与自身线粒体分布和ΔΨm的差异。结果D3冷冻优质胚胎组线粒体均匀分布于每个卵裂球核周围,染色均匀;而质量较差胚胎组线粒体不均匀的分布在卵裂球内,总体染色不均匀;在D5冷冻优质胚胎组线粒体主要密集在内细胞团核周围较宽的区域,染色较深且均匀,而在质量较差胚胎组线粒体主要分散在内细胞团核周围,染色较浅且不均匀。D3冷冻优质胚胎组中胚胎线粒体ΔΨm高于质量较差胚胎组[(2.660±0.039)vs.(1.800±0.034),P<0.05],D5冷冻优质胚胎组中胚胎线粒体ΔΨm明显高于质量较差胚胎组[(3.400±0.061)vs.(2.420±0.056),P<0.05];结论胚胎线粒体分布能够反映胚胎发育潜能,ΔΨm反映胚胎活性,质量较好的胚胎线粒体分布均匀且ΔΨm较高。Objective To investigate the association of morphological typing of human embryos and blastocysts during cleavage with mitochondrial distribution and mitochondrial membrane potential(ΔΨm).Methods In vitro fertilization-embryo transfer(IVF-ET)was performed in the reproductive center due to female bilateral fallopian tube obstruction.After the healthy fetus was successfully delivered,the frozen cleavage embryos and blastocysts were voluntarily provided for scientific research.After frozen embryos thawing recovery,according to the different embryonic development period,all the embryos were divided into points D3 frozen embryos(cleavage stage embryo)and D5 frozen embryos group(blastocysts),and then based on the criteria of frozen embryos,frozen cleavage stage embryos were divided into high-quality embryo and poor-quality embryo group.Fluorescent probe JC-10 was used to detect the mitochondrial membrane potential of embryos,and the differences of mitochondrial membrane potential between embryos at different stages and embryos of different quality at the same stage and their own were compared.Results In D3 high-quality embryo group,mitochondria were evenly distributed around each blastomere nucleus and stained evenly.In the poor-quality embryo group,mitochondria were unevenly distributed in the blastomere,and the overall staining was uneven.In D5 high-quality embryo group,mitochondria were mainly concentrated in a wide area around the inner cell mass nucleus,with deep and uniform staining;while in poor-quality embryo group,mitochondria were mainly scattered around the inner cell mass nucleus,with shallow and uneven staining.MitochondriaΔΨm of high-quality embryos in D3 frozen embryo group was higher than that in the poor-quality embryo group(2.600±0.039 vs.1.800±0.034,P<0.03),and the mitochondriaΔΨm of high-quality embryos in the D5 frozen embryo group was significantly higher than that of poor-quality embryos(3.400±0.061 vs.2.420±0.056,P<0.02).Conclusion Embryonic mitochondriaΔΨm reflects the activity of e
