四种消毒剂对猪圆环Ⅱ型病毒核酸降解效果研究

摘　　要：为了评估进口过硫酸氢钾复合物粉(A)、国产过硫酸氢钾复合物粉(B)、烧碱(C)、戊二醛(D)在不同场景对猪圆环Ⅱ型病毒核酸降解差异性,将病毒液与粪便残渣混合后分别涂抹在室外车辆轮胎(场景1)、室外露天地面(场景2)、室内地面(场景3)、室内地面尿液中(场景4)。室外组按照当时室外温度进场操作。室内组设置三个温度(4℃、25℃、37℃)。上述4个场景分别在不同时间点取样,用QPCR检测病毒核酸相对含量。结果发现,在消毒剂消杀后,PCV2核酸转阴时间:场景1,消毒剂A、B(7h)、消毒剂D(5 h);场景2,消毒剂A、B、D(>7h);场景3,消毒剂A(>5h)、消毒剂B(≥7h);消毒剂D(>5 h);场景4,消毒剂A(>3 h)、消毒剂B(>3 h)、消毒剂C(>7h)、消毒剂D(>3 h)。效果较好的消毒剂(推荐):场景1用D消毒剂;场景2用A消毒剂;场景3中春秋季用A消毒剂,夏季用D消毒剂;场景4中用B消毒剂。A In order to evaluate the difference of nucleic acid degradation of porcine circovirus type Ⅱ by imported potassium persulfate complex powder(A),domestic potassium persulfate complex powder(B),caustic soda(C),and glutaraldehyde(D)in different scenarios,porcine circovirus type Ⅱ virus liquid and fecal residue were mixed and smeared respectively in outdoor vehicle tires(Scenario 1),outdoor ground(Scenario 2),indoor ground(Scenario 3),and indoor ground urine(Scenario 4).The outdoor groups shall enter the site for operation according to the outdoor temperature at that time.The indoor groups were set with three temperatures,namely 4℃,25℃ and 37℃.The above four scenarios were sampled at different time points,and the relative content of viral nucleic acid was detected by QPCR.The results showed that after the disinfectant disinfection and sterilization,the time of PCV2nucleic acid turning negative:Scenario 1,disinfectant A,B(7 h),disinfectant D(5 h);Scenario 2:Disinfectants A,B,(>7 h);Scenario 3,disinfectant A(>5h),disinfectant B(≥7 h);Disinfectant D(>5h);Scenario 4,disinfectant A(>3h),disinfectant B(>3 h),disinfectant C(>7 h),disinfectant D(>3h).Disinfectant with good effect(recommended):glutaraldehyde is used for disinfection in Scenario 1;Scene 2:Use imported potassium persulfate complex powder for disinfection;In Scenario 3,in spring and autumn,the imported potassium persulfate complex powder is used for disinfection,and in summer,glutaraldehyde is used for disinfection;In scenario 4,domestic potassium persulfate complex powder was used for disinfection.
